Homogeneous time-resolved fluoroimmunoassay(HTRF)has been widely applied in the disease diagnosis,biomarker,PCR,and kinase etc.It is one of the analysis method which has the high sensitivity and good prospect.The synthesis of bifunctional chelating agent is the important and difficult of this research.In this dissertation,the work are as follows considering the technology demand.Three chelate intermediates and A kind of chelator Eu3+-cryptate have been successfully synthesized: ethyl-2(2,6-dimethylpyridine-4-yloxy)acetate,ethyl-2(2,6-dibromomethylpyridine-4-yloxy)acetate,2,6-dibromomethylpyridine-4-carboxylic ester and Eu3+-pyridine(COOH)-bipyridine-bipyridine.The products were characterized by fourier transform infrared spectrometer(FTIR),nuclear magnetic resonance(NMR),mass spectrometry(MS).The fluorescence spectroscopic properties of the structure of the chelator was researched.The most important advantage of lanthanide fluorescene chelate is that the wide excitation spectrum,the narrow emission spectrum and the high quantum yield,the stokes shift is 298 nm and the fluorescence lifetime is 828μs.It can fully satisfy the need of HTRF and lay a foundation for its growth.
