Time-resolved fluoroimmunoassay (TRFIA) is an ultrasenstive analytical method, instead of radioimmunoassay, it has been widely used in diagnostics and clinical analyses, iatrology, life sciences. Using the chelator of rare-earth ion labeling Ag or Ab, the high sensitivity of rare-earth ion detection can be obtained with time-resolved fluorometry, because of the fluorescent character of chelator: exceptional fluorescence life and large Stokes'shift. When measuring fluorescence, using befitting time and measuring wave, the disturbance of background can be eliminated effectively.There exist two directions: DELFIA and DSLFIA in the field of TRFIA research. DELFIA is named by using enhanced-solution when measuring. High sensitivity can be obtained with DELFIA, but the operation is inconvenient. DSLFIA is widely concerned because of operating conveniently and the appling foreground in DNA hybridization assays and immunoassay fluorescence image. The chelator used in DSLFIA should not only have the bifunctional properties: conjuncted with protein and chelated with rare-earth ion, but also transfer the energy to rare-earth ion. An ideal chelator is the key technology of DSLFIA.In this dissertation, there was a big paragraph talked about the synthesis of two bipyridinery intermediates 4,4′-bi(p-nitroethynylbenzene)-6,6′-dimethyl-2,2′-bipyridine and Tetraethyl 2,2′,2′′,2′′′-[(4,4′-Dibromo-2,2′-bipyridine-6,6′-diylbis(methylenenitrilo)]tetrakis (acetate), which applied to solid-phase time-resolved fluoroimmunoassay. The constructions of all products were proved by DSC, IR, element analysis, 1H-NMR. The condition of the synthesis was researched. We studied the reacted temperature, reacted time, catalyst amount, quomodo of adding material, propirtion of material, and improved the method of synthesis, and enhanced the yield.
