L-isoleucine methyl ester hydrochloride and linalool are sex pheromone of diablo gill beetle,(Z)-3-hexenyl acetate and(E)-2-hexenyl acetate are plant volatiles which have synergistic effect on the sex pheromone of diablo gill beetle.These four compounds are used as the target drugs to prepare gas synergistic sustained-release gels.The quality standard draft system of synergistic sustained-release gels were established.It is divided into five parts.In the first part,the hygroscopicity and volatility of four main components of crude drugs were studied,the results showed that the hygroscopicity and volatility of four main components of crude drugs were strong.So crude drugs could be prepared into synergistic sustained-release gels.The principal component 1 was used as the represent to determine the gel basic substance was carboxymethyl cellulose sodium,the solvent was ethanol,and their proportional range and drug ratio range were confirmed.The second part is prescription and process optimization of the diablo gill beetle sex pheromone synergistic sustained-release gels.Drug residue amount was used as indicator to optimize the prescription of four diablo gill beetle sex pheromone synergistic sustained-release gels with orthogonal experiment.The verification results show that the sustained release effect of optimal prescriptions were excellent in 30 D.The structures of the sustained release gels were characterized by infrared chromatography and the results showed that the excipients structure of the drug did not be affected by the chemical.In the third part,the release dates of diablo gill beetle sex pheromone synergistic sustained-release gels were fitted with 6 release models(zero-order、level 1、Higuchi、Ritger-Peppas、Baker-Lonsdale and Hixon-Crowell model).The results showed that drugs release of diablo gill beetle sex pheromone synergistic sustained-release gels respectively conform to Baker-Lonsdale、 Hixon-Crowell 、Hixon-Crowell and level 1 models.The drugs release mechanism were diffusion and corrosion.In the fourth part,the quality standards of diablo gill beetle sex pheromone synergistic sustained-release gels were established.According to the preparation quality standard of "China Pharmacopoeia" 2015,character,identification,pH,viscosity,particle size and other projects of four synergistic sustained-release gels were investigated.The content analysis methods of four synergistic sustained-release gels were established using ultraviolet spectrophotometry and gas chromatography.The methodological study results showed that two kinds of content analysis method is simple and accurate,its precision,stability and recovery were good.Ultraviolet spectrophotometric is suitable for industrial production and gas chromatography is suitable for the precision analysis.In the fifth part,the stability of diablo gill beetle sex pheromone synergistic sustained-release gels were studied.The results showed that the appearance,pH and the content of four kinds of synergistic sustained-release gels were normal in stress testing,accelerated testing and long-term testing and were stable for 9 months under low temperature.In summary,four diablo gill beetle sex pheromone synergistic sustained-release gels were developed.The appearance of smooth,stable properties,pH value and particle size were in line with the requirements of the Pharmacopoeia.The analysis methods are simple and accurate,and these gels are suitable for industrial production.
