In this study, attention was paid to sex pheromone of diablo gill beetle, to preparethe sustained-release preparations of volatile substances. At first, preparationtechnology and release rule of the above sustained-release preparations were studied,and then its quality system was builted, what`s more, its release performance wereverified through field trap experiment. Meanwhile, release mechanism of the abovesustained-release preparations was investigated which could provide a reliabletheoretical foundation for its development.The following several parts were included in this study: The volatile of Linalooland L-isoleucine methyl ester hydrochloride which were two parts of sex pheromonewere investigated in the first part, the results show that L-isoleucine methyl esterhydrochloride has less volatile,the vocs was only3%when put it in anhydrous ethanol48hours.The vocs of Linalool was50%in48hours, and it completely volatile in90hours,and the vocs of Linalool sustained-release preparations was45%in90hours,sothe target durg can be prepared into sustained-release preparations.Preparation technology of diablo gill beetle sex pheromone sustained-releasepreparations were studied in the second part, common medicinal polymer materialswere chosed as the slow-release carrier,and orthogonal experiment was used for thechoice of optimum preparation process. The result clearly indicates: the best xcipientswas β-CDs or starch, process condition was given priority to drug and excipients ratioof1:1. Verification tests shows that the relationship between residual and time of pastesustained-release preparations using β-CDs as excipients was Y=0.0442X2-2.8181X+98.241, R=0.9994;and the granular sustained-release preparations wasY=0.1163X2-5.0964X+99.11,R=0.9992. The optimized two kinds of sustained-release preparations both have regular slow-release performance.The quality standard of the sustained-release preparations of diablo gill beetle sexpheromone was built in the third part. The general content of preparation qualitystandard in2010edition of the People`s Republic of China pharmacopoeia was referenced to build the character, the identification, the pH, the particle size and theviscosity.The two active ingredients have similar maximum absorption wavelength inUV, so use the maximum absorption wavelength of the two active ingredients as thecontent determination wavelength of the compound ingredients. The results show thatthe precision and stability of the method were well, recovery of paste controlled-release preparations was99.88%, RSD was2.38%; recovery of granular controlled-release preparations was100.2%, RSD was2.23%, and the excipients didn`t haveaffect on the content determination of drugs, the method was simple, so this methodcan be used as the content determination of the slow-release preparations.The stability of two kinds of sustained-release dosage forms was investigated inthe fourth part. The results show that light and high humidity had little influence at thecontent, high temperature had a great effect on it. Under the condition of40℃, after10days the color of granular turned yellow and the content decreased by0.4%; thecolor of the paste was not change, but content decreased by0.5%. Long-term testingresults show that the two kinds of sustained-release preparations were stable during thesix months in room temperature.In the fifth part, we verify the slow-release effect by field trap test, the resultsindicates the trap effect of two kinds of controlled-release dosage forms wereconsistent with the experimental data, the trap time of paste can be up to30days,granule can be up to20days.Two kinds of sustained-release dosage forms of volatile substances weresuccessfully prepared in this experiment and their determination method and qualitysystem was builted. Stablity of the above two dosage forms was studied, and valuablerelease parameters of the two above dosage forms were also proposed. Application ofsex pheromone which was energy-saving and environmentally friendly was providedbased on the field application data.
