| Fluorescence analysis has the some advantages,such as of small amount of sampling,high sensitivity and good reproducibility,it has been developed rapidly both in theory and in application,and was applied in the fields of chemistry,biology,medicine,agriculture and environment protection.Hydrazine is an important chemical material,but it is harmful to human.So,it is particularly vital to develop a reliable,sensitive,selective,and convenient analytical method for the detection of hydrazine,but there are few reports on the use of fluorescent probes for hydrazine detection.In this paper,three fluorescent probes for the detection of hydrazine were designed and synthesized taking fluorescein and coumarin as fluorophore,and the detection of hydrazine in the physiological environment was also studied.At the same time,a new NO2-probe was prepared using modified coumarin.The main content follows:Chapter 1: The Introduction,mainly introduce the basic concept of the fluorescent probe,detection mechanism,and the related research of hydrazine detection,and our design ideas was provided;Chapter 2: Fluorescent probes 1,2 of hydrazine detection based on fluorescein were synthesized through introducing the ester bond,C=C group to the skeleton;the test method to hydrazine was realized taking advantages of the strong nucleophilicity of hydrazine,the relative ultraviolet and fluorescence properties of the probes were measured in HEPES buffer solution(pH = 7.4).The results indicated that Probe 2 possesses good selectivity and specificity to N2H4,suggesting it is a reactive fluorescent probe molecule with favorable performance.Furthermore,the corresponding reaction mechanism was speculated by MS.Chapter 3: Fluorescent probe 3 was designed and prepared by bonding 2-fluoro-4-nitro benzoic group to coumarin core,and the ultraviolet and fluorescence properties of the reaction between Probe 3 and N2H4 were tested in HEPES buffer solution(pH = 7.4).The results showed that the detection of Probe 3 to N2H4 was fulfilled due to the nucleophilic attack of N2H4 to F and C=C successively,the protecting group 2-fluoro-4-nitro benzoic was removed and core coumarin was released,thus,it is confirmed that the Probe 3 is a good reactive molecule to hydrazine.Chapter 4: a new NO2-probe was obtained using connecting NH2-group to 6-position of coumarin.Based on Sandmeyer Reaction mechanism,the probe has good properties to detect NO2-ion under acidic condition.The synthesis route of probe is simple,the response time is rapid,and the selectivity is fine. |
