This research investigated the intervention effects of High amylose corn resistant starch, chitosan oligosaccharide and their compound powder on metabolic disorder which induced by high-surcose high-fat diet, through measuring body weight, blood-glucose, lipid metabolism related indexes, oxidive stress related indexes, liver function and explored that if resistant statch and chitosan oligosaccharide can exert synergistic effect. Then measured the expression quantity of glucose manipulation and lipid metabolism related genes by using RT-PCR method, to analysis the potential molecular mechanisms how it intervening metabolism disorder.Resistant starch was used as ingredient and mixed with COS, then adopted spray-dying method to prepare RS-COS compound podwer, the physicochemical properties was measured by the SEM, laser diffraction analyzer and FTIR spectrometer. The results showed that the property of compounds were obviously differtent to raw RS. RS granules were covered by COS and adhered with each other, leading to the particle size of the compound become bigger than the raw starch granules. besides, compound showed higher oil adsorption rate and lower digestion rate than raw RS. at the digestion endpoint,compared with RS, the glucose release quantity of Compound decreased 9.02%.Experimental rats were divided into 5 groups, including normal diet group,high-surcose high-fat diet group and 3 different administration groups, administration groups were treated with COS(0.3g/d-per rat), .RS( 1.2g/d-per rat) and compound(1.5g/d-per rat), normal diet group and high-surcose high-fat diet groups' rats treated with same volume distilled water. After 6 weeks,measured rats' body weight,body fat, liver mass, blood glucose level, serum insulin level, serum lipid level, feces lipid profile,oxidative stress and liver function related index, made liver tissue pathological slides. The results indicated that RS and COS could exert synergistic effect after cross-linking reaction,to significantly decrease high-surcose high fat diet rats' body weight and body fat ratio;decrease serum insulin level, improve insulin resistance; decrease serum TC, TG, LDL-C levels, increase lipid excreation in feces; decrease MDA level, increase T-AOC, T-SOD and GSH-Px level, improve oxidive stress state; lower liver index and serum ALT, AST and AKP level, inhibit Fatty degeneration of hepatocyte.Through RT-PCR method, the expression quantity of blood glucose manipulate and lipid metabolism related genes were measured, the results showed that diet administration could facilitate the conversion from blood glucose to hepatic glycogen, inhabit glucose synthesis and gluconeogenesis processes, inhabit fatty acid, triglyceride, chelosterol bio-synthesis, facilitate fat ?-oxidation.Given the overall results, the intervention effect of the compound was better than RS or COS administrated alone, indicated RS and COS complex can exert additive effects to inhibit metabolism disorder progress which induced by high-surcose high-fat diet.
