Acarbose is an efficient medicine for curing the type II diabetes.It has stronger inhibition effect to the intestinal digestive enzyme,and occupied the marker owing to its well affection and low virulence,the demand is increasing year by year.In our laboratory,a strain Actinoplanes.utahensis ZJB-08196 with higher synthesize ability of acarbose was screened,this study was aimed at enhancing the acarbose titer and reducing the concentration of component C.The main work and the results are as follows:The detection methods for acarbose fermentation parameters were established.The optimum inoculation amount and fermentation volume were 10%and 30 ml in 250 ml-flask,respectively,and we made the curves of seed growth and fermentation process.Some kinds of representative aminocyclitol compounds on the acarbose fermentation were investigated,and it resulted that only C7N aminocyclitol compounds could increase the acarbose production.We established an suitable fed-batch fermentation strategy:fed the mixture consist of 6 g/1 glucose,14 g/1 maltose,and 9 g/1 soybean meal twice at 72 h and 96 h.It was found that validamycin A could enhance the production of acarbose,adopted the above fed-batch fermentation strategy with 0.08 g/1 validamycin A addition at the beginning of fermentation,as a result the acarbose titer reached to 5602 mg/1,increased by 57%compared with the contrast,and the component C induced by 54 simultaneously;All of valienamine,validamine and validamycins lysate ha enhancement effect on acarbose titer,The adding of 20 mg/1 validamine into t medium with fed in 72 h and 96 h obtain the maximum acarbose 6606 mg/l,wh was increased by 90%compared with the control,and the byproduct component C was decreased by 64%simultaneously.Through further research on the fermentation parameters,it was deduced that appropriate amount of validamycin A and validamine could enhance the consumption of carbohydrate,the growth of cell and the accumulation of acarbose,and inhibit the generation of byproduct component C.This study provided an effective control method for acarbose fermentation.Validamycin A,valienamine and validamine are likely to be used as precursor directly and indirectly involved in the biosynthesis of acarbose.Adding these substances not only enhanced the accumulation of acarbose,but also inhibited the generation of byproduct component C,which will bring more conveniences to product separation.Our next step will investigate the mechanism of this phenomenon to provide a theoretical basis for industrial application.
