In this paper, the techniques of fermentation , separation and purification of acarbose were studied.Acarbose is an inhibitor of α-glucosidase, currently used for diabetes mellitus cure. In this paper, the acarbose producer-Actinoplanes sp.AC-5 was used as a starting strain(with a acarbose titre of 411.6μg/ml). A high yield acarbose producing strain AC17 with a titre of 1416μg/ml was obtained by treatment of mycelia with supersonic, followed by the mutagenesis of UV light.With strain AC17 the components of slant medium, the slant cultivation time, the slant preservation time and methods were investigated. An better slant medium and optimal slant cultivation time as well as better slant preservation conditions were obtained. Then, a seed medium and cultivation conditions suitable for industrial production were obtained after the optimization of components of seed medium and seed cultivation conditions (including the pH , the filling volum of seed medium, seed cultivation time).Based on above results, the fermentation medium and cultivation conditions were investigated and the uniform design method was used in the optimization of fermentation medium. The techniques of fed-batch culture were also investigated in this paper, with this techniques acarbose fermentation titre increased from 1416 μg/ml to 2700 μg/ml, basically reached the level of industrial production.In addition, the separation and purification techniques of acarbose were also studied ,a procedure of extraction and purification of acarbose was set up.
