Chelerythrine(CHE)is a bioactivity-rich alkaloid that is resistant to cancer,pathogens and Oncomelania.It is less abundant in plants and has difficulty to be separated.Therefore,the efficient separation of chelerythrine has been attached much more attention.In this study,a new type of core-shell-shell magnetic molecular imprinting polymers was synthesized.The adsorption kinetics and thermodynamics of the polymer microspheres were studied,and the efficient separation of CHE was carried out.Furthermore,the mechanism of interaction between CHE and key hole limpet hemocyanin(KLH)was also discussed.It provided a useful reference for the development of CHE.The main experimental content is as follows:(1)Chelerythrine-Magnetic Molecular Imprinting Polymers(CHE-MMIPs)were synthesized by free radical polymerization with tetraethylpyridine as functional monomer and CHE as template.And the appearance of the characterization was carried out.It could be seen that the microspheres have a narrow particle size distribution(about 240nm),good magnetic response(>40emu/g),and good monodispersity.(2)The adsorption efficiency of CHE-MMIPs was investigated.The results showed that the maximum adsorption capacity of CHE-MMIPs was 12 mg/g(6 h),and the adsorption process was in accordance with Langmuir isothermal adsorption model and pseudo-second-order kinetic equation.(3)The quenching mechanism between chelerythrine(CHE)and key hole limpet hemocyanin(KLH)was investigated by fluorescence spectra,synchronous fluorescence spectra and molecular docking technology.These results revealed the intrinsic fluorescence of KLH was strongly quenched by CHE through a static quenching procedure.At 293 K,298K,303 K,the binding constants of CHE and KLH were 4.72 × 105,5.06 × 105,1.01 × 106 L / mol,respectively.The thermodynamic parameters ?G298K,?H298K and ?S298K of the system are-32.54 k J/mol,-58.78 k J/ mol,and-88.05 J/ mol·K,respectively.The van der Waals forces and hydrogen bond formation were played major roles in the binding process.The specific position,number of sites and binding affinity of CHE and KLH were obtained by molecular docking technique.It was found that the Cu site in KLH was not a potential binding site for CHE and KLH.
