Thais Roeding is a common edible snail in Fujian sea,but it can be used as medicine.However,due to the regional distribution of natural distribution,there are few studies on its bioactive substances at home and abroad.Therefore,the effect and mechanism of Thais Roeding The selection of natural medicine is of great significance.In this study,lychee sprouts were used as extraction materials,and the active components were extracted by organic solvent extraction method.The cells were isolated and purified by thin layer chromatography,silica gel column chromatography and preparative liquid chromatography.The cell proliferation was detected by cell proliferation and necrosis Detection,mitochondrial membrane potential detection and PARP protein clipping and other means to analyze the cell level of anti-cancer activity,to explain its anti-cancer mechanism.The results showed that it could inhibit the proliferation of breast cancer cells MCF-7 and QGY-7703 cells by the MTS method.The results showed that the concentration of QFY-7703 was significantly inhibited by the method of silica gel column chromatography.At 40 ?g/ml,the survival rates of the two cancer cells were significantly decreased(<56%).EA-2 could induce PARP protein cleavage in hepatocellular carcinoma cell line QGY-7703 and increase cell viability after adding pan-Caspase inhibitor.At the same time,EA-2 could significantly decrease the mitochondrial membrane potential of MCF-7 cells,and the apoptotic changes of cell morphology were observed.The number of apoptotic cells increased significantly.The IC50 of EA-2 to MCF-7 and QGY-7703 was 28 ?g/ml and 46 ?g/ml,respectively,and its inhibitory effect on cancer cells was mainly through apoptosis.Finally,the active components were further separated and purified by silica gel column chromatography and preparative high performance liquid chromatography to obtain high purity compounds(98%).The structure was identified by nuclear magnetic resonance and mass spectrometry,which proved that it was 6-bromoisatin.Most of the natural medicine comes from terrestrial plants,because of its safety,the characteristics of small side effects by the people of widespread concern.In this study,21 kinds of plant-derived compounds were screened for antitumor activity,and tried to provide theoretical help for the study of plant-derived natural drugs.21 compounds were used to treat human bladder cancer cells T24 and KU-19-19.Drugs capable of inhibiting cell proliferation were screened out.Then,the cells were treated by cell proliferation,apoptosis and necrosis,mitochondrial membrane potential.Activity for further analysis.Screening results showed that cucurbitacin B had significant inhibitory effect on T24 cells.By further analysis,cucurbitacin B could significantly change the morphology of cancer cells,and the cytoplasm was screened by Hoechst 33342 and PI staining.Nuclear fragmentation,and the production of apoptotic bodies,the number of apoptotic cells increased with the drug concentration.The decrease of mitochondrial membrane potential also proved the anti-tumor effect of cucurbitacin B in the way of apoptosis.
