Saded soy isoflavones powder as substrate,object with the enteric bacteria LG-Q2 from vegetarian in feces.namely the quail chicken Enterococcus(Enterococcus.Gallinarum)for work.Research the preparation and biological activity evaluation of equol.Using single factor experiment,orthogonal experiment and response surface method,on the basis of previous work,examines the growth curve of the work,different carbon sources on the influence of the equol.The results showed that the growth rule for 37 ? under anaerobic conditions,develop into index after 6 h,smooth into the growth period after 24h,as the decline phaseafter 39 h,livingbacterium number is 1.34*108.Glucose as the optimal carbon sources,the Transformability of equol is 2.21 ?g/mg.The optimal fermentation condition,concentration of glucose in the fermented liquid of 5.00 g/L,pH 7.02,NaCl concentration is 3.93 g/L,the Transformability of equol is 2.58?g/mg,prove model was established.Added the different concentration gradient of glucose,maltose,lactose,sucrose and xylan different carbon source,to determine the best carbon source.Results showed that glucose as the optimal carbon sources,At this time,the transformability of equol is 2.21?g/mg.With carbon source,pH and osmotic pressure for single factor,equol production as evaluation indexes,Through the response surface experiment investigation of transformability of equol with LG-Q2,Best conditions for glucose concentration was 5.00 g/L,pH 7.02,NaCl concentration was 3.93 g/L,the biggest forecast of transformability of equol was 2.59?g/mg,Actual measured of transformability of equol is 2.58?g/mg,Active carbon decoloring,polyamide resin as adsorbent for purification processing,the freeze-drying preparation of equol.Decoloring treatment and purification processing of culture,Experiment with bleaching time,dosage and decoloring temperature for single factor,Decolorization rate as the evaluation indexes,Through the test to determine the best single factor,in order to do the orthogonal experiment,results show that in the process of decolorization effect on decolorization rate the biggest factor for bleaching time.the best decoloring conditions was temperature of 50 ?,decoloring dose of 2 g,decoloring time of 1.5 h.Did with the purification of adsorption,elution steps by the elution rate,dosage of adsorbent and eluent for single factor,Finally get the transformability of equol as the evaluation index,Do orthogonal experiment to get the best condition of purification processResults show that the influence for transformability of equol was the dose of eluent,the best purification conditions was elution dose of 40 mL,elution rate of 0.6 mL/min,absorption dose of 2 g.Doing the recovery test with the Standards of equol,show the yield is 0.25%,the recovery was 78.38%.In feeding way for the mice with standard of equol,sample of equol,soy isoflavone and Saline,to take blood from mice eyes,do Determination of kits with TG,TC,BCA,LDL-C,HDL-C and NEFA.In order to get the equol effects on mice blood fat.Resulted show that the standard of equol,sample of equol,soy isoflavone all can effects on mice blood fat,the largest,effect was standard of equol.At the end of the experiment was carried out on the test of male mice cervical dislocation executed,Take the testicular tissue in the body of HE staining,observe the Testicular tissue structure of the slice with microscope,The results was the sample of equol does not affect in male mice testicular tissue.
