Compared with the traditional drug delivery process,the drug release system has the advantages of reducing the number of times of administration,reducing the side effects of drugs on the body,prolonging the time of drugs in vivo and so on.The use of biodegradable biopolymer materials made of drug-loaded fibers,with a large surface area,more pore structure,good mechanical properties and other characteristics,in the drug delivery,tissue engineering,biomedical field development is of great significance.Therefore,in order to further expand the application of fiber in drug sustained-release carrier,the development of a drug-loaded fiber material with sustained-release properties has certain significance.Electrostatic spinning technology is a simple and easy method for preparing fibers.The prepared fiber has the characteristics of size control,rich porosity,high specific surface area and so on.It is widely used in drug release materials,tissue engineering,Adsorption experiments and other fields.Chitosan(CS),as a carrier of natural alkaline polysaccharides,has the characteristics of non-toxic and non-irritating,good biocompatibility and biodegradability,and has attracted much attention as drug carrier material.However,due to its solubility,polycation characteristics and other issues make CS in the electrospinning process is limited,pure CS difficult to direct electrospinning fibers.Because CS is difficult to form fiber as a difficult electrospinning material,adding a synthetic polymer which is easy to be electrospun and has biocompatibility and biodegradability,with good mechanical properties,can improve the specific surface area of the fiber and then blended in the CS solution,and finally the preparation of polymer/chitosan/inorganic composite fibers,used to improve the fibers specific surface area,drug loading and so on.In this paper is intended to prepare a drug-loaded material.The natural macromolecule polymer CS,synthetic polymer and inorganic orthosilicate(TEOS)were mixed,using the electrospinning technology,the use of the characteristics of the material,by adjusting the concentration of the material,the ratio of conditions such as optimization,to obtain the best proportion,synthesis of a certain specific surface area,a certain porosity of the drug-loaded fiber membrane,and to explore the effect of spinning parameters on the spinning effect.Adding the drug to the best proportion of the spinning solution to obtain the drug-loaded composite fiber membrane,the effects of different contents of the drug on the composite fiber membrane and the sustained release properties of the fiber were discussed.The details are as follows:1.Synthetic fiber membrane were synthesized by electrospinning using polyamide(PA)/chitosan(CS)/ethyl orthosilicate(TEOS)as raw material.The optimum ratio of 1:0.13:0.67 was determined by adjusting the parameters in the spinning blends.Different concentrations of ofloxacin(OFLO)were added to the spinning solution with the best concentration in vitro.When the drug content is 0.2%,the slow release performance is the best,cumulative release was 71%.2.Replace the polymer is polyvinyl alcohol(PVA).PVA/chitosan(CS)/ethyl orthosilicate(TEOS)was used as raw material to synthesize composite fiber membrane by electrospinning.The optimal ratio was 1:043:1.2 by adjusting the parameters of the spinning blends.The fiber membrane had a specific surface area(7.805 m2/g)and a narrow pore size structure.In the best proportion of the spinning solution to add different concentrations of ofloxacin(OFLO)drugs,in vitro release study found that the fiber membrane has a sustained release effect,cumulative release was 69%.
