Capillary electrophoresis?CE?,which uses capillary separation channel and high-voltage DC electric field as the driving force,is an analytical technique with efficient and rapid separation for the analytes.In recent years,the high thermal efficiency and small diameter capillary tube is used in CE to improve the separation quality.It is widely used in life sciences,food science,clinical medicine,analytical chemistry field because of its high separation efficiency,low sample consumption,rapid analysis,etc.Electrochemiluminescence?ECL?is a combination of electrochemical means and chemiluminescence method.It is widely used in the field of biological,environmental,food,medicine,pharmacy,clinical,chemical analysis due to the advantages of high sensitivity,simple equipment,easy operation,good controllability and selectivity.The biogenic amines are found in meat,aquatic products,wine,milk and fruits and vegetables and other foods.As aquatic products are rich in amino acids,so their bodies contain a variety of biogenic amines,and the corruption and stored improperly are likely to improve the content of biogenic amines significantly.Low content of biogenic amines are considered to be a physiologically active substance,but excessive levels of biogenic amines will cause toxic it.So,it is especially important to establish an efficient and rapid method for the detection of biogenic amines in aquatic product.CE-ECL has many advantages to analyze the biogenic amines in aquatic product,including CE's efficient,fast and ECL's sensitivity,reproducibility.At the same time,highstabilityandluminousefficiencyRu?bpy?32+isselectedtoact electrochemiluminescence system.The purpose of this study was to establish a new method for the separation and detection of biogenic amines in aquatic products.The results of this research are listed in the following three aspects:1.A CE-ECL coupled with use Ru?bpy?32+as electrochemiluminescence system was established to separate and detect histamine of squid.Results indicated that:under the optimized conditions,such as detection potential at 1.20 V,photomultiplier tube voltage at 800 V,5 mmol/L Ru?bpy?32+?pH=7.0?,sample injection at 8 s×12 kV,separation voltage at 11 kV,50 mmol/L of phosphate buffer at pH 7.0.Under the maximal condition,the ECL intensity was linear with the concentration of phenyl ethylamine in the range of 140?g/mL.The linear equation for Y=1.73x+9.71,correlation coefficient R=0.9981 with detection limit?S/N=3?of 0.06?mol/L.Determination of phenyl ethylamine in fish sauce,The RSD of peak height and migration time were 0.08%and 1.40%,Recovery is 102.19108.08%.2.CE-ECL was first applied to detect tryptamine in pomfret.Under the optimal condition:trytamine dihydrochloride was linear with concentration of trytamine dihydrochloride in the range of 18 mmol/L.The linear equation for I=20.649 c+29.839,correlation coefficient R=0.9996?n=8?with detection limit?S/N=3?of 0.06 mmol/L.3.In this research,a novel method is built for determination and separation of histamine in shredded squid by CE-ECL and Ru?bpy?32+was used to be electrochemiluminescence system.Results indicated that:under the optimal conditions,such as detection potential at 1.15V,5mmol/L Ru?bpy?32+?pH=7.0?,sample injection at 8s×10kV,separation voltage at 18kV,50mmol/L of phosphate buffer at pH 3.4.Under its optimal conditions,the ECL intensity was linear with concentration of histamine dihydrochloride,the linear equation for Y=1.42x-30.83,correlation coefficient R=0.9981?n=6?with detection limit?S/N=3?of 0.468?mol/L.The method used to determinate histamine in the squid silk samples.The RSD of peak height and migration time were 0.43%and 0.29%,recovery is 100.32106.55%.4.Oysters are the desirable seafood in Xiamen,but there is a few report about its content of biogenic amines.In this experiment,CE-ECL Ru?bpy?32+system was used to separate and detect biogenic amines?putrescine,histamine,tyramine,phenylethylamine and spermidine?in fresh oysters.The changes of biogenic amines content at different storage conditions were studied and analyzed.The experimental results showed that the fresh oysters spermidine higher levels and low levels of Tyramine and Histamine.Levels of various biogenic amines with storage time have different growth.Store at 0°C for 4 days,the content of biogenic amines changed little,with varying degrees of growth from the first four days,which Putrescine from 18.6 mg/kg increase to28.3 mg/kg,Spermidine from 45.6 mg/kg increase to 68.5 mg/kg.4°C under conditions,from the next day after the biogenic amine content started to grow,four days after the putrescine and spermidine rapid growth to,putrescine to increase to 136.8 mg/kg from 19.4 mg/kg,spermidine increased from 46.7 mg/kg to 119.7 mg/kg,the accumulation of biogenic amines rate significantly higher than 0°C.
