Purification And Identification Of The Procyanidins From The Red Grape Seed Of Hetian And The Wine Of Musalais

Red grape of Hetian is the main producing in the Hetian,Xinjiang province.Due to the particularity of the region.which may lead to some of nutrient composition is different from other grapes.Its biological activity is very widely,mainly in the antioxidant,free radical scavenging,antitumor,prevent cardiovascular disease and so on.Musalais is a special wine in Xinjiang.Which materials are from the Red grape of Hetian.In the paper,we have studied the extraction process of procyanidins and optimized from the grape seed.Then refine the extracts by the resin method and optimized the conditions of the purification.The products were grape procyanidins and Musalais procyanidins after resin managed.Which were further purified by SephadexLH-20,eluted with 50% methanol,methanol and 70% acetone.The tube number was abscissa and the absorbance was the ordinate as the elution curve.According to the elution curve,The combined components were analyzed using LC-MS.All of the results are as follows:The single factor test results of extraction of procyanidins in the grape seed as follows: acetone extractant,temperature was 60℃;extraction times was 90 min,pH value was 4,material ratio was 1:20 The optimum extraction conditions were as follows: solvent of 62.10% ethanol;extraction times was 86.50 min;temperature was 53.65 ℃;pH value was 3.68;grape seed to ratio of solVEnt was 1:20.Under the condition of the theoretical maximum withdrawal rate is 24.65%,the verification experiment which was carried out to get the extraction yield was 24.02%.Through the selection of the four kinds of resin,AB-8、NKA、ADS-17、ADS-21.Based on the static adsorption curve and the static desorption curve.The AB-8 was selected to purified last.The optimal loading and elution conditions as follows: adsorption speed was 1.5 BV/h;concentration was 15mg/mL.loading volume was 5 BV;eluent was 60% ethanol;adsorption speed was 2 BV/h;adsorption pH was 6;desorption speed was 2 BV/h,desorption volume was 4 BV;The purity of procyanidins is 88.89% after purification.The LC-MS analysis results showed that the purified procyanidins from the grape seed and Musalais after AB-8 resin.Grape seed contain gallic acid,Procyanidin B1,Procyanidin B2,(+)-Catechin,(-)-Epcatechin;Musalais contain Procyanidin B2 and Oligomer mixture.The results of further purification of Sephadex LH-20 showed that there were more kinds of procyanidins in the grape seed after purification,and the content of oligomers was higher than Musalais.
