Colored rice is a kind of special rice which is becoming more and more attractive with its unique flavor,the colorful color and the rich nutrition.Nutrients which includes a variety of vitamins and trace elements are beneficial to human body.In addition,the polyphenols compounds endowed the colored rice with strong antioxidation activity.At the same time,the studies of colored rice also have reported have the function of reducing weight,but the inhibition mechanism of generation and accumulation of lipid is uncertain.The purpose of the study is to investigate the polyphenols compounds in seven different kinds of colored rice extracts,which has strong antioxidation activity and the ability of scavenging free radicals and potential activity of anti-obesity.These results will be used to determine whether the colored rice has the effect of treatment or prevention of obesity.This experiment mainly includes three parts.The first part was to extract colored rice,determine the content of total phenols and its antioxidant activity by using a variety of free radical systems such as DPPH free radicals,ABTS+ free radicals and the super oxygen free radical O2-ability.The results showed that the variety of the higher contents of total phenol and flavonoids,have the stronger antioxidant ability,embodied in its ability to scavenge the free radicals.The flavonoids in colored rice has certain contribution on total phenol,both have great contribution to its antioxidant activity.The second part was using ultraviolet spectrophotometry for the full wavelength scanning,and high performance liquid chromatography(HPLC)method to analysis the component of polyphenols in extracts.Results showed that polyphenols are the material basis of its antioxidant,mainly gallic acid,catechin,ferulic acid,vanillic acid and caffeic acid,et al.The third part was using C3H10T1/2-clone8 mesenchymal stem cells in vitro studies to determine whether the colored rice extracts can inhibit lipid formation process of differentiation.For differentiation cells,using oil red O staining to quantitate the lipid droplets,using the realtime fluorescent quantitative polymerase chain reaction(Realtime-PCR)assay to measure lipid metabolism related genes(PPARγ,C/EBPα,Fabp4,AdipoQ,Plin1)mRNA expression which was used to analyze the differentiation degree of the lipid.Results showed that after dealing with the extracts,the lipid metabolism related genes expression had down-regulated when compared with positive control group.To detect the PPARγ protein contents by using the western blot method,PPAR gamma is peroxidase proliferation agent activated receptor,it was classed as a nuclear receptor transcription factor that is essential for adipogenic differentiation.The results showed that PPAR gamma protein expressed the largest in positive control group(ADM),the treatment group GER has the lowest expresion,RER is followed by GER.The results showed that different colored rice extracts have different degrees of inhibition on lipid differentiation process,which means that the colored rice extracts can regulate the express of the PPAR gamma protein to inhibit the generation and accumulation of lipid. |
