Curcuma.Longa L.is a traditional Chinese medicine;curcuminoids,represented by curcumin,demethoxycurcumin and bisdemethoxycurcumin,are the main active ingredients of Curcuma Longa L..JH1 is a new natural product of Curcuma Longa L.with strong anti-inflammatory activity.The LC-MS analysis method established in this study is of high precision,good reproducibility,high accuracy and stability.Results of cluster analysis showed that the JH1 content of Curcuma Longa L.originated from Sichuan and Chongqing was significantly higher than that in other areas.In view of the fact that curcumioids have many significant biological activities,they have strong potential to be developed for new antitumor drug system,but their water solubility and bioavailability hindered the clinical application and new drugs development.Tea polyphenols and curcuminoids are both the natural phenolic ingredients derived from plant extracts,they not only have similarity in chemical structure but both have strong antioxidant effect,which has significant effect on inhibiting the proliferation of tumor cells.This study adopted the film-ultrasonic method to embed curcuminoids and tea polyphenols in solid lipid nanoparticle,first established a method for the qualitative and quantitative analyses of CTP-SLNs by using the technology of HPLC-DAD-ESI/MSn-a complete method for compound preparation;Moreover,the study preliminarily evaluated the anti-tumor activity of CTP-SLNs in vitro.Mass Spectrometric Analysis of LC/MS has shown that eight active monomers of curcuminoids and tea polyphenols remained stable in the preparation.HPLC Fingerprint analysis showed that among 20 batches of compound nano preparation,17 batches have similarity of more than 99%.All batches of compound nano preparation composition were basically stable.During the storage period,the CTP-SLNs diameter has small fluctuation,it showed that the properties of nanoparticle remain stable in the six storage months,without any degradation or aggregation.The result of MTT experiment showed that the combination of curcuminoids and tea polyphenols had synergistic inhibitory effect on the proliferation of tumor cells HepG2 and CT-26;and the combination of solid lipid nanoparticles had stronger inhibitory effect than curcumin,EGCG,curcuminoids,tea polyphenols,or the mixture of curcuminoids and tea polyphenols alone(P < 0.05),so it embodied the superiority of solid lipid nanoparticle when used as drug delivery carrier,it improved the bioavailability of curcuminoids and tea polyphenols,thus raised the lethality of cancer cells and the effectiveness of the drugs.
