Abiotic stress including drought,high temperature,low temperature,high salt and biological stress including pests and diseases will seriously effect the growth and development of plants.The perception and transmission of external signals are mediated through the activation of different phosphorylation pathway by protein kinase.It is known that the plant stress resistance,hormone signal transduction,growth and development regulation are related with the protein kinase.Lectin receptor-like kinases(LecRKs)and casein Kinases(CK)are two common protein kinases with important physiological and biochemical functions in plants.The physiological function of Arabidopsis LecRK-ConA gene in LecRLK family and CKB1 gene in CK family was studied in the present study.Three primer PCR methods were used to identify the homozygous T-DNA mutants of LecRK-ConA gene.The analysis of Cis-elements in promoter showed that LecRK-ConA gene had multiple drought,light and GA responsive elements.The results of real-time quantitati’ve PCR showed that the expression of LecRK-ConA gene had organ difference and it had the strongest expression in germinating seeds.The expression of LecRK-ConA gene was inhibited under ABA treatment in Arabidopsis。The seed germination rate of lecrk-cona was lower than that of wild type under NaCl and mannitol treatments.Furthermore,lecrk-cona mutants flowered early compared to Col-0 plants.The promoter region of the casein kinase CKB1 gene was analyzed and it was found that the promoter region contained a number of cis acting elements associated with light response and stress response.Two mutants of CKB1 gene were screened by"three primers".Two homozygous T-DNA insertion mutants of the CKB1 gene was obtained.The CKB1 expression vector was successfully constructed by the gateway method,and the transgenic plants were obtained.The wild type and mutant and overexpression lines as materials,by The two-dimensional gel electrophoresis proteomics analysis showed that there were some differential proteins in the wild type,deletion mutants and overexpression lines.There different some proteins were found in wild type and mutants,these differential proteins were associated with photosynthesis and stress response signal transduction by retrieving database analysis.The above results showed that the LecRK-ConA and CKB1 protein kinase genes played the important roles in the growth and development of plants.It provided important references for further study of their physiological functions and their applications. |
