Based on optical trapping effect,optical tweezers is a tool to capture and operate micro particles.The optical tweezers could capture particles like biological cells without contact,and it has good transmission characteristics when adopting infrared light,thus the damage on the particle is reduced and this characteristic makes optical tweezers widely used in the field of biology chemical and medicine.What's more,optical tweezers can be also applied in other fields as accuracy of the manipulation and measurement and sensor.Due to the rapidly development of optical fiber technology,it will be applied in broader fields.Firstly,the paper,researches the way to calibrate the optical trapping stiffness by using thermal motion analysis method and makes numerical simulation to the motion of the particles captured by fiber optical tweezers using Monte-Carlo numerical modeling method.The effect caused by the exposure time,the particle radius,the initial position and the temperature has been also argued in this paper which provides a theoretical basis and foundation to the calibration the optical trapping stiffness.Secondly,this paper puts forward the key technology of the overall design of the fiber optical tweezers system.The paper analysis the demand of the fiber optical tweezers system and completes the overall design.Then the paper presents the design and implementation of optical,mechanical and electrical design,and has the control observation and measurement of fiber optical tweezers system realized,which laid the foundation for the popularization and application of optical fiber optical tweezers system.Finally,due to the fiber optical tweezers system that is set up particle capture experiment is completed and optical trapping stiffness is marked out.The capture and the operation of polystyrene spheres which is of different sizes is realized by using cone type optical fiber probe.Meanwhile transverse optical trapping stiffness and longitudinal stiffness of optical tweezers was calibrated,which provides the basis for the subsequent measurement of optical trapping force.
