Tettigonioidea is the second largest group in the order Orthoptera,and there are more than 7,000 species in the world,belonging to more than 1,200 genera.There is no doubt that the actual number should be far more than this number.In this paper,DNA barcode technology was used for the first time to classify insects of Tettigonioidea.Main work inc1 ude : 1)Collect samples and extract DNA;2)PCR amplification of target fragments using related amplification primers;3)Sequencing of amplified target fragments;4)Comparison and analysis of sequences to obtain 658 bp sequence.5)using the R language,ABGD,BIN,jMOTU,RESL to analyze the results;6)the research sample information submitted to the DNA barcode database BOLD system in order to establish the initial DNA barcode database of Tettigonioidea.Thus rapidly identifing the species of Tettigonioidea will come true.The results include:1.A total of 1169 Tettigonioidae COI sequences were studied,belonging to 6 subfamilies of 23 genera and 116 species.A total of 963 COI Sequences of Phaneropteridae was included,belonging to 3 subfamilies of 29 genera of 152 species.There both was no significant "Barcoding gap" in the interspecies of COI gene.2.Regardless of BIN,RESL,jMOTU,the three methods of dividing the OTUs are far more than the morphological division of the species indicating that the diversity of species increased,and Tettigonioidea's division also need to conduct a deeper study for there may be a large number of cryptic species.For example,the Gampsocleis carinata is divided into three OTUs,and the Ducetia japonica is divided into three OTUs.3.The results of Hexacentrus showed that except for Hexacentrus japonicus,the barcode gap existed in the other species.However,there were 3 BINs in Hexacentrus japonicus(BOLD: ACX8110;BOLD: ACD8277;BOLD: ACD 8278).It is recored there exist 6 species of Hexacentrus.The Taxon ID tree indicated that there are 8 BINs(BOLD's barcode index numbers),which reveal the existence of cryptic species in Hexacentrus japonicus Based the DNA barcode technique.The application of DNA barcodes has accelerated the research process of biodiversity,and it can be seen that molecular diversity can increase the diversity of Hexacentrus.
