The Identification And Expression Analysis Of LncRNA-Ptma

With the development and perfection of biological technology(high throughput sequencing technology),a growing number of LncRNAs have been found.But by far the LncRNAs have been researched to get their function are just few.LncRNA research is still in primary stage.This research uses RNA-seq data combined with bioinformatics technology to screen and get a new gene,which is located in 1 chromosome(chr1)in mice,the LncRNA is named as LncRNA-Ptma because of its target gene-Ptma.First of all,I get the 3'end full-length of by 3'RACE.The sequencing results show that the sequence has a significant polyA tail sequence and AATAA termination sequence.Using the 5 'RACE experiment method to obtain a band with a length of 423 bp,the sequencing results show that the sequence has a significant peak of transcription initiation site.At the same time,the experimental method of Northern is used to verify the number and size of the transcript,it includes 652 bp and one transcript.Finally,less than 200 nt ORF,negative CSF and the result of the cell's nuclear position indicate that LncRNAPtma is a long non-coding RNA.Next I focus on the LncRNA-Ptma gene expression patterns.Combined with in-situ hybridization,the result reveals that the LncRNA-Ptma is expressed in mouse embryos and organizations,and brain tissue owns the highest expression level.Quantitative PCR result in E15.5 is consistent with the in-situ hybridization.It also shows a dynamic expression model of LncRNA-Ptma gene in the brain,liver,lung tissues in mice embryonic developmental different periods,and the peaks are concentrated expression in E15.5.It indicates that the gene may be involved in the development of middle or late-stage organs in mouse embryos.Above all,the research obtains a mouse embryonic development related new geneLncRNA-Ptma,and successfully obtains the full-length of LncRNA-Ptma and transcription information.Then I get the expression pattern of LncRNA-Ptma and the preliminary exploration of the LncRNA-Ptma gene function.All the results suggest that the LncRNA-Ptma plays a role in the development of different organizations and organs.It aslo provide security foundation for the subsequent knockout mice model.
