The Human Genome Project has concluded, but its successful completion has increased, rather than decreased, the need for high-throughput DNA sequencing technologies. DNA sequencing is one of the important means of biological research.The development of accurate and high-throughput DNA sequencing methods has a strong impact on biomedical research. The DNA sequencing by synthesis (SBS)approach based on chemical synthesis has shown great promise as a new sequencing platform, with particular progress reported recently.In this article, we design and synthesize a acid-sensitive linker (A1) , a reactive oxygen species responsive thioketal linker (B1) and a class of reversible terminator(la-lc) based on cleavable disulfide linker. All the synthesized products are characterized by 1HNMR?31PNMR?HRMS. And we continually conducted a experiment of DNA sequencing for one of the synthesized disulfide reversible terminator, which obtains good effect of extension and cleavage in the course of experiment of extension and shows great prospect of application.
