With research on DNA sequencing technology deepening, DNA sequencing technology has been rapid development. In people's daily lives, the application of DNA sequencing technology is more and more widely. It plays an important role in promoting the development of clinical medicine and basic biology. However, the current DNA sequencing technology still can not meet people's requirements for low-cost, large-scale parallel sequencing. Thus, to explore new DNA sequencing technology still has important practical significance.In DNA sequencing by synthesis, as the core of the cycle reversible terminal, the linkers plays a direct and decisive role to the sequencing efficiency. Thus, firstly, this paper has designed and synthesized a benzaldehyde-type linker T006, cyclopentanone-type linker T016, acetone fork-type linker T011, p-methoxy-type linker T022. After filtering out the right linkers, this paper has also synthesized the modified deoxy-uridine acid dUTP-NH2 and deoxycytidine acid dCTP-NH2. And 7-deaza-7-iodo-2'-deoxyguanosine (G010) were synthesized. Then the linkers reacts with the modified deoxynucleotide acid and fluorescein. Ultimately, We can get the acid-sensitive cycle reversible terminal T036, which can be used to the DNA sequencing by synthesis.
