Researchs On The Extract Of Effective Substances And Activity Of Gardenia Jasminoides Var. Radicans

The extract of effective substances of Gardenia jasminoides var.radicans and its efficacy are studied in this paper,which aims to develop the application of gardenia resources in health foods.The subcritical methods using petroleum ether,95%-ethanol,and water was employed to extract the effective substances of Gardenia jasminoides,and the extraction temperature,extraction time,liquid-solid ratio was explored to establish optimum conditions with the yield as index.Based on corresponding regression model,the optimum extraction parameters of PEE were as follows: liquid-solid ratio of 6:1,extraction time of 15 mins,and temperature of 100?,with the yield of Gardenia petroleum ether extraction of 24.32%.Meanwhile,the optimum extraction parameters of ET were as follows: liquid-solid ratio of 6:1,extraction time of 18 mins,and temperature of 110 ?.The yield of ET was 35.32%.Effective component was detected by reagent reaction and functional component was detected by high performance liquid chromatography?HPLC?.Results showed that total phenol,total flavonoids,total iridoid,total carotenoids,total crocin of ET were 0.71%,2.16%,7.95%,1.24%,7.95%,respectively.And the WE contents were 0.29%,0.42%,5.35%,0.19%,0.38% each,the effective components in petroleum ether extraction were 0.33%,1.02%,0.95%,0.03%,1.35%,separately.The gardenia acid,geniposide,geniposide and crocin-1,crocin-2 and crocetin in the three extracts were determined by HPLC analysis,which demonstrated that the gardenia acid,in three kinds of content is extremely low,almost not detected,while geniposide is the most.For efficacy,the antioxidant activity,decreasing blood sugar and neuroprotection effect of three extracts were researched.The antioxidant activity was determined by establishing DPPH generation system,and half inhibitory concentration?IC50?of Vc and PEE,ET,WE,and of which the WE showed the best ability.The effect of extracts on ?-glucosidase was researched after,and the IC50 of acarbose,PEE,ET,WE and of which the ET showed the best effacts.For neuroprotection effect,the inhibiting ACh E activity results showed that the ET has the best effects.In cell experiment,the concentration of A?25-35 damaging PC₁₂ cell was 40 ?mol/L,which was obtained through the establishment of A?25-35 injury model,and in this condition,results suggested the three extracts had protective effect in 100 ?g/m L,and the difference of which was significant?p<0.05?.
