With the increasing number of new drugs and the combination of drug prescriptions,a lot of potential drug interactions induce adverse reactions appear constantly, which not only gives the patients an extra burden, even a serious threat to life safety of patients. At present, the study of adverse drug reactions still pay more attention to a single drug, and drug interaction induced adverse reactions remained far from adequate.. In view of drug interactions, drug interactions have become a major hazard to the safety of drug use. the adverse reactions caused by multi drug interactions were depth analyzed in this research,the purpose of the research is to quickly and effectively detect the risk signal between DDI and ADR in combination of drugs, and to promote the use of multiple drugs in the clinical practice, in order to reduce the incidence of adverse drug reactions.Blood glucose increased and Blood glucose decreased were selected as research objects,we use the traditional baseline model and data from 2013 to 2015 in the FDA Adverse Event Reporting System (FAERS),to study the relationship between DDI and ADR risk signals of two dimensional drug combination. Because the baseline model is not suitable for high dimensional (2 to 5 dimensional) drug combination, the mixed logistic regression model is used to solve the problem of high - dimensional drug.The signal of two dimensional drug combination interaction was screened by the baseline model. (including multiplicative model, additive model),then preceded statistical test,wo obtain the exact risk signals of drug combination. the mixed logistic regression model is comprised of a constant risk model and a dose-response risk model. Using empirical Bayes estimation method, local false discovery rate as the judgment condition,we can effectively detect drug combinations.whose risk signal between DDI and ADR is high.We use baseline model to screen the 2 dimensional drug combinations of blood glucose increased and blood glucose decreased, 1251 suspicious ADR signals were detected. There are 103 cases of secondary exact risk signals were found after statistical test.High-dimensional drug combinations of two kinds of adverse reactions are treated by the mixed logistic regression model as follows:first,on the basis of regression analysis, we observed the risk of adverse reactions increases as the drug interaction dimension increases;second, We calculate the local false discovery rate of the drug combination.and sort them, when the dimension was 2 to 4, 10 groups of drug combinations whose ADR signal was high were screened out. When the dimension is 5, the number of drug combinations is changed to 5.The result show that the two models used in this research can effectively detect the risk signal of DDI and ADR in drug combination. furthermore, which with strong ADR signal were screened out. and the potential drug safety problems can be found from a large number of adverse drug reaction data,so the subject has practical significance.
