Circular RNA(circRNA)is a cyclic,non-coding RNA transcript that is structurally different from common linear RNAs.Circulating RNAs are widely present in various fields of life,including eukaryotes,bacteria,archaea and viruses.Recent studies have shown that the formation mechanism and function of circular RNAs are diverse,such as exon cyclization,intron cyclization,bacterial genome cyclization and so on.A variety of mechanisms contribute to the diversity of circular RNAs,with a variety of functions,it can be used as miRNA sponge,RBP sponge,translation protein.The ring RNA is present in a variety of forms in the genomic,viral or virus-like genomes of the exons of the eukaryotic exon,such as the tRNA and rRNA introns of the archaea,in the form of eukaryotes.The exonuclease recognizes the 3 'end,so it is possible to resist the degradation of the exonuclease.Stable,conserved,mature circular RNA is abundantly present in the cytoplasm.Can be used as a miRNA sponge,a typical example of miR-7 sponge.Combined with RBP synthesis of RNA-protein synthesis,or mediating gene transcription.As the circRNA has antagonistic ability,has a strong stability in the differentiation of mature cells can be slowly accumulated,so it can be used as biomolecule markers in clinical,scientific research.In addition,studies have shown that ring RNA may be associated with microRNA(miRNA)molecules,exogenous virus can inhibit the function of these molecules.Ring RNA may have great biological significance,the current study of circular RNA is still lacking,especially the intron source of circular RNA.In this study.Arabidopsis thaliana as the object of study,according to the characteristics of RNA such as circular RNA,intron branches,intron source of circular RNA recognition and analysis.In this study,the experimental group was treated with exonuclease(RnaseR)and no control group.No 8 samples were repeated 4 times in 2 groups.The samples were then subjected to high-throughput sequencing.According to the characteristics of the branch points,the branch points are identified in the intron and the characteristics of the branch points are explored by analyzing the conservation of the branch points.The gene expression levels of 8 samples were calculated and the differences between the two groups were analyzed by clustering and dispersing the gene expression levels.In addition,the expression of the intron is calculated by the normalized expression formula.Screening intron-derived circular RNAs based on intron expression levels were selected to represent a significantly up-regulated circular RNA.Comparison of gene and intron differences in the two groups,focusing on the analysis of introns.The cyclic RNA was analyzed by length and other characteristics.This paper makes a detailed and careful analysis of the characteristics and experimental data of the circular RNA,and makes the following results:1)identified 4080 branch points;2)found that the branch point of the nucleotide is highly conserved,most in the 3' shear site upstream 19-37nt region and other characteristics;3)9227 intron-derived circular RNAs were identified from Arabidopsis thaliana(control and experimental)by RnaseR-treated high-throughput sequence data.4)The circular RNA was significantly up-regulated in 172 experimental groups compared with the control group by differential analysis.5)The length of the circular RNA is generally 400bp,longer than the normal intron length;In summary,this topic identified the nesting and intron source RNA of Arabidopsis thaliana,and further analyzed its characteristics.Hope to be able to help after the study of Arabidopsis thaliana or circular RNA.
