WRKY is one significant member of the transcription factor family in the plant.It plays a very important role in transcription regulation during the process of plant growth and development.We obtain 20 Do WRKY transcription factors?conserved regions baseing on the data of.the Dendrobium officinale protocorms' transcriptome.We further study the expression of the DoWRKY6 gene in different years,different parts and tissue culture seedings of the Dendrobium officinale.We conduct phylogenetic analysis and the analysis of conserved sequence in the conserved region of 20 WRKY genic sequences.These members have at least one conserved domain composed by the "WRKYGQK" amino acid residues.We find that there are significant difference in the 20 WRKY genetic affinity with building the phylogenetic tree.Through the bioinformatics analysis I obtained the protein sequence characteristics of the DoWRKY6 in the Dendrobium officinale.The DoWRKY6 has a complete ORF genic sequence which is 1017bp long encoding 339 amino acids with one WRKY domain and a zinc finger structure of CX5C23HX1H.It belongs the II WRKY factors.It can also specially bind with the W-box.We predict that the protein molecular formular for the DoWRKY6 is C1641H2556N454O530S13 through the Protparam.The DoWRKY6's relative molecular weight is 37541.8 whose theoretical isoelectric point is 4.86.DoWRKY6 genic GRAVY is-0.756.It also is a hydrophobic protein.The techology of Real-time quantitative PCR is used to obtain the characteristic expression in different organs and different years of Dendrobium officinale.The DoWRKY6 mainly expresses in the stem.And the expression greatly reduces along with the increasing of the growth years.DoWRKY6 expresses the most in the stem nearby the bud,and expresses the secondly in the basal stem.The result showed that DoWRKY6 gene has significantregulation on the growth and development of the stem in the Dendrobium officinale.
