Eupolyphaga sinensisWalker,a Chinese traditional medicinal insect.There are many microorganisms in the insect gut.Intestinal microbiota play an important role in the life activities of the host.In order to study the intestinal microbiota diversity of Eupolyphaga sinensisWalker.Illumina Hiseq sequencing technique was used to measure the sequence of 16 S rDNA V4 variable region of intestinal microbiota in 3 samples of Eupolyphage sinensisWalker.The traditional cultivate method was used to analyzethe diversity of aerobic and facultative anaerobic bacteria in Eupolyphaga sinensisWalker,s gut.30 phyla,187 families and 201 genera were detected in three samples through sequencing.Bacteroidetes?Proteobacteria and Firmicutes were dominant phyla.Porphy--romonadaceae,,Sphingomonadaceae,Lachnospiraceae,Xanthomonadaceae,Enterobacter iaceae,Rikenellaceae,Burkholderiaceae,Pseudomonadaceae,Chitinophagaceae,Sphingob acteriaceae,Bacteroidaceae,Bacillacea and Methanomassiliicoccaceae this 14 families werepredominantatfamilylevel.CandidatusAzobacteroIde,Parabacteroides,Burkhol--deria,Ruminococcu,Kaistobacter,Pseudomonas,Bacillus and vadinCA11 were predomi--nant at genus leve.Seven aerobic and facultative anaerobic bacteria strains were obtained from the intestinal contents of Eupolyphaga sinensisWalkerby means of pure cultivation.The strains were classified to Serratia,Corynebacterium,Lysinibacillus,Oceanobacillus,Ba--cillus,Pseudomonas and Rhodococcus.The Serratia strain can produce red pigment.The pigment was preliminarily identified as prodigiosin and the production of red pigment was affected by temperature.The fermentation of this red pigment producing strain have no inhibitory effects onStaphylococcus aureus,Escherichia coli,Salmonella typhosa,Pseudomonas aeruginosa and Salmonella Pullorum.Blattabacterium is common exist in the fat body of blattodea as endosymbiont.This symbiotic bacteria can support almost all of the essential amino acids for blattodea.In order to research the fat body endosymbionts of Eupolyphaga sinensisWalker.Illumina Hiseq sequencing technique was used to analyze the PCR products about 16 S rDNA V4 region from Eupolyphaga sinensisWalker,sfat body tissue.The result show there were Blattabacterium in the fat body of Eupolyphaga sinensis Walker.And other 178 genera of bacteria with lower abundance such asAlistipes were also detected.
