In this paper,the Drosophila melanogaster as the object of study,we constructedthe pAAV-AlstR-IRES-EGFP plasmid,and injected the plasmid in the Drosophila blastoderm by microinjection technique.we constructed AlstR transgenic Drosophila,and provedby fluorescence detection,PCR verification and the development process of observation.Further to study the influence of AlstR overexpression on Drosophila social behavior,we established the climbing model and the modelexploring space,comparing the difference of social behaviorbetween the transgenic and wild type Drosophila.At the same time,the life and survival experiments help in Drosophila as a screening platform for anti-aging and anti aging Old related drug.The research results show that:(1)The carrier report gene enhanced green fluorescent protein(EGFP)expression detected that plasmid successfully transfected and expressed.Microinjection of plasmid led to the short development time of the eggs of Drosophila melanogaster.Alst R gene has a certain degree of influence on the development of Drosophila melanogaster from larva to pupa.The effect of AlstR gene on the developmental process of male and female Drosophila melanogaster was different,and the probability of male Drosophila melanogaster to develop into adult was significantly decreased by micro-injection of exogenous genes.(2)Compared with the wild type,the development of motor related ability of AlstR Drosophila melanogaster was slow,but its ability to maintain a long time.Meanwhile,the intelligence and social communication ability of 5 week old AlstR flies were significantly higher than that of wild type Drosophila at the same survival time.Therefore,AlstR gene significantly improved the intelligence and social communication ability of Drosophila melanogaster.(3)AlstR inhibited the maturation of male Drosophila melanogaster in larval stage and improved the longevity of Drosophila melanogaster when the fruit flies develop into adults.The gap in life expectancy between the two sexes is reduced.And Alst R could significantly improve the survival ability of male Drosophila melanogaster under UV radiation.At thesame time,the establishment of Drosophila melanogaster UV radiation model is beneficial to the screening of anti aging and anti-aging related drugs.
