The continuous development of science and technology,has prormoted the modern development biology into the microcosmic field.The invention and development of different kinds of experimental technology,has played great promoting roles in the basic life sciences research.The invention and application of EM,which is seen as an important observation technology,have the meaning of milepost for the development of life science.With the appearance of TEM,scientists have obtained great achievement in the research of biological ultrastructure and the analysis of the dynamic biological action.Great relationship has been verified between the structure and function of skin/skin appendages.To explore the reactivity and structural change of skin/skin appendages under specific physiological or pathological condition,the TEM technology was used here to observe the ultrastructure of skin/skin appendages from two different model organisms,mouse and chicken,aiming at obtaining morphological results under specific conditions,which would provide some evidence for the research of development biology.In this paper,the main experiments with transmission electron microscopy are following three aspects:1)Ultrastructure observations during the process of chicken feather branch development,including various locations of chicken feathers(anterior and posterior,proximal and distal,barb,not the branch region).The experiment results show that,there are changes of filipodia on basal cells during the branching process.Disturbance of the development process of chicken feathers with small molecules S2188(the inhibitor of Notch),feathers were collected two days later.It was shown that the filipodia of epidermal cells on rachis were shortened or even disappeared,which indicated that Notch signaling may play a role in this process.2)To explore the effect of chemotherapeutic agents on the cell morphology,chemotherapeutic agents were used to treat chickens,and the feathers were collected at different time points.Through ultrastructural observations,the apoptosis was mainly studied.The results showed that apoptosis did not appear at the rachis,but there are some apoptosis at the barb area.3)To explore the effect of ion radiation on skin cell morphology,the mice foot skin was exposed to ionizing radiation(40Gy),Samples were collected at a specific point in time.The results showed that cell apoptosis does not occur in the process,while the spacing between cells was broadened.Also,the thickness of the epidermis were lowered.However,in the ninth day after irradiation,the skin began to recover.Through these experiments,TEM has great value in clinical development and scientific issues,which will provide a basis at the feather developmental problems,chemotherapy and radiation damage mechanisms based on above results.
