With the development of science and technology,the ability of human beings to explore the earth has been further enhanced.Marine resources will become the focus of contention in various countries.There are abundant marine resources,including marine biological resources,mineral resources,energy resources and microbial resources.Different exploration technologies are needed for different resources.Microbial resources are widely used in the field of life science and have important medicinal value.Because deep sea microorganisms survive in high pressure,low temperature and light environment,in order to fully understand the physical and chemical characteristics of deep-sea microorganisms,we usually research the original samples of deep-sea microorganisms.It has a high demand for the holding pressure sampling technology,the Microbial pressure maintaining and culturing technology and the Microbial pressure maintaining and transferring technology.At home and abroad,there has been a lot of research and Application on the technology of Microbial pressure maintaining and sampling and Microbial pressure maintaining and culturing in deep sea microorganisms.However,the technology of Microbial pressure maintaining and transferring is still in its infancy.The main technical difficulties in the Microbial pressure maintaining and transferring technology is how to ensure high pressure,how to carry out effective sample transfer in high pressure environment,and how to reduce the pressure fluctuation because of sample transfer or leakage caused by other reasons.This paper will study these issues.This thesis includes six chapters as following.In Chapter 1,we first introduce the background of the research on the Microbial pressure maintaining and transferring of deep-sea microorganisms,and then introduce the current situation of the research on the technology of Microbial pressure maintaining and transferring in foreign countries and china.Because of the deep sea resources for the purpose of this paper,the overall design scheme of the Microbial pressure maintaining and transferring technology of deep-sea microorganisms is given.According to the technical difficulties of the overall scheme,the main contents of the three aspects of the mechanical,electronic control and pressure maintenance are put forward;In chapter 2,the working principle of the Microbial pressure maintaining and transferring device is designed,including the design of the pressure maintaining unit and the design of the sub sampling unit.Then,the design and calculation of the pressure bearing parts,such as the pressure holding cylinder,the screw rod and the supporting plate,are carried out.In addition,the finite element analysis software is used to simulate and optimize the structure of the cylinder and the support plate;In chapter 3,we firstly analyzes the reasons which may lead to pressure fluctuation of the microbial pressure maintaining and transferring system,such as slide,leakage and valve open and close,and assessed their influence on the microbial pressure maintaining and transferring system,and gives the measures to avoid.After referencing to the commonly used design methods of pressure maintaining circuit and combining with the structure characteristics of the microbial pressure maintaining and transferring device,we designed the pressure maintaining system in which A one-way valve and accumulator are used as the pressure maintaining components,and the pressure maintaining circuit is properly supplemented by the hydraulic pump when the accumulator can not keep enough pressure.In chapter 4,firstly,the control requirements of the microbial Microbial pressure maintaining and transferring system are analyzed,and the requirements of real-time,accuracy and reliability of the electronic control system are determined.In order to solve these problems,this paper designs the control scheme of the host computer and the slave computer.And then,the implementation of the upper computer,the data acquisition of the slave computer,the communication protocol between the host computer and the slave computer are designed in detail;In chapter 5,combining with the research results of the above three chapters,such as the mechanical structure,electronic control software and pressure maintenance,we successfully created a microbial pressure maintaining and transferring system prototype.We experimentally illustrated the pressure holding ability of the device,and verifide the correctness of the principle of mechanical structure and the real-time control performance of the control software through the sampling experiments,which lays a foundation for further research;The sixth chapter is the summary and prospect,summarizes the research results of the deep sea microbial pressure retaining device,and makes a prospect of the technology of the more advanced microbial pressure maintaining and transferring system.
