| In the field of life science research,Caenorhabditis elegans as a model organism classic material is used by more and more scientific experimenters.The count of elegans is the basis of almost every test of nematode.Fast and accurate counting and classification of nematodes are important to research radiation damage and extraction of genes.The traditional method of counting and classification of nematodes is the use of a fluorescent microscope,but fluorescence microscope has some shortcomings,for example bulky,expensive.This paper designs detection system of nematode counting and classification based on microfluidic chip.The detection system has some advantages for example:a small size,high detection efficiency,low sample consumption,and low price.The advantage of this designed system is counting nematode while still achieving classfying nematode.The main contents of this paper are as follows:(1)The detection principle of based microfluidic chip count and classification of nematodes is determined.Respectively RPS differential counting method and laser-induced fluorescence taxonomy.Difference RPS counting method is capable of counting individual particles or sample.Laser Induced Fluorescence is that some of the fluorescent protein within excitation light is capable of producing fluorescent.(2)The detecting system is designed based on the above two methods.The detecting system consists of RPS counting module,laser-induced fluorescence modules,power modules,filter amplifier module,signal acquisition displaying and memorying module,the mechanical structure of the module.RPS differential counting module and laser-induced fluorescence classification module is the core module of detection systems,this paper designs a small dark mechanical structure providing external support for the normal operation of two core modules and designs eight kinds of detection channels of microfluidic chip,and choose the best by experimental comparison.Because the difference RPS signals and fluorescence signals are weak signals,in order to obtaining better signal,this article designs a weak signal amplification and filtering circuit and simulates.(3)The above modules are tested.Ensuring each module working properly,experimental about N2 wild-type and AM141 nematodes is begun,and the experimental results are compared with fluorescent microscope,it proves that the result of article detection is correct and reliable.The detection limitation of nematode width is about 10 um. |
