Male sterility (MS) is one of the important foundation of crop heterosis utilization, is the hotspot of plant developmental biology research. Hybrid rice made great contribution to China's grain production since 1976 popularized? Hybrid rice breeding depends on the selection and application of excellent male sterile ? The nuclei sterile, as the ptcl, can't be used in the production widely, which is not affected by the temperature and light, due to lack of maintenance line? Because of the homology between OsPTCl and AtMS1 and the advantages arabidopsis thaliana as a model plant, our will indicate the genetic networks of PTC1, through screening and identifying the suppressor of msl in Arabidopsis, then studying its homologous genes in Rice? That will lay a foundation for creating the nucleus sterile maintainer line The main results are as follows:1. Because of the infertility of mutant msl, in order to get the hidden suppressor of msl in the M1 generation, we introduce the system of Glucocorticoid Receptor to created a transgenic msl plants carrier pro MS1:msl:GR. Through it we can control the fertility of mutation. That had laid the foundation for mutagenesis work.2. The seeds of msl-GR were treated in EMS, then obtaining 5 fertile plants in M1 generation, named 16,61,89,98,111 respectively. We take a cytology observation about silique, anther and pollen of these plants. On the one hand, these plants were sterility compared with msl, displayed long silique, filled seed and mature pollen grains in the anther. On the other hand, these plants are slightly different from wild type in the degree of viable of pollen grains.3. We were confirmed the background of each plant the results show that the five plants are homozygous msl genotype. In the hybrid F2 segregation population of 98 and wild type, the plants without ProMS1:msl:GR still viable, excluding the the sterility caused by segments of transgenic escape into the nuclear.4. In the hybrid F2 segregation population of 98 and wild type(Ler), we selected 30 plants randomly that were sterility and homozygous msl for sequencing, taking a rapid gene location by the MutMap. Combining the way of mutation and the gene expression patterns, we were determined the candidate gene AT1G21470, named Smsl-98. For the moment, there are no reports about Smsl-98. We carried on the preliminary analysis using bioinformatics technology. The protein sequence alignment results show that Smsl-98 is CCCH zinc finger family protein, and the domain are very conservative in many plant species. The prediction of expression spectrum shows that Smsl-98 had a higher expression in the stem leaf, sepals and mature pollen et al.5. In the end, we has taken a tentative analysis about the homologous genes in rice LOC_Os06g42020.1(named OsSmsl-98), and prospecting the application in hybrid rice breeding.
