The study of crustaceans decapoda sperm non-condensed nucleus experienced an earlier think that the bare existence of ite generic material DNA and no histone bound to it,then the further research that the basic protein were found associated to DNA and formed the special nucleosomes, the latest result revealed that during the spermiogenesis histones apparently leave the nucleus and migrate to the acrosomal granule,this process also refer to the histone epigenetic modification,which have an influence on the decondesed chromatin.This article takes the histone variant H3.3 replaces histone H3 thereby giving an influence on the activety of chromatin and the linker histone H1 variant take assistance on the substitution of core histonne as a window for which had been studied comprehensively in epigenetics.Through the technology of RACE,the full length gene h3.3 and h1 histone variant were coloned then their codon region were constructed in the expression vector and would express in prokaryotic expression system. The fusion protein H3.3 was pufried by nickel noclumn and used to detect the specificity antibody that had been purchased from Abcam. As histone variant H1,we measured its relative quantitative with usual H1 in matured sperm in E. sinensis by qPCR.To this we studied whether H3.3 exist in mature sperm and the expression quantity of H1 variant impacted on sperm non-condensed nuclus.The results show that h3.3 was not expressed in the mature crab. For the relative quantitative H1 and its variation,both of them exist in the mature sperm and acroding to special proportion take part in the structure of nucleosome,which influence the non-condensed nucleus chromatin of E. sinensis.
