This paper consists of two parts, which are as follows:PART ONE: reviews1. Following the deep research on Sox gene family, more and more new members and new founction of Sox gene were continnously revealed, species proved having Sox genes were increasing.In this paper,the recent research of Sox gene family were reviewed including the identification, naming, classification and founction of Sox gene.The regulation founction of Sox gene in developing process is specially introduced, such as sex determination, embryogenesis, neural system development, lens development, neural crest development, chondrogenesis, haemopoesis, oogenesis, heart and blood vessels development .This paper can provide leads for further study of Sox gene.PART TWO: studies1. Using a pair of degenerate primers based on the conservative region, HMG-box, of Human SRY gene, tow different fragments of Sox gene, ESSox3 and ESSox22 were amplified from female and male Eriocheir sinensis, the sequence results indicated that ESSox3 and ESSox22 shown high homology to Human SOX genes, and the identities to human SOX genes in DNA sequence and amino acid sequence are 84%,85% and 97%,81%, respectively.It might be concluded that Sox gene was highly conservative in phylogenesis. The results provided molecular data for the further study of sex-determining mechanism of Eriocheir sinensis and evolution of Sox gene.2. The Sox gene expression analysis of different tissues from Eriocheir sinensis was studied by using RT-PCR. An expression was observed in testis in male ,heart in both male and female , no expression in ovary in female, muscle in both male and female, the expression of Sox gene bore distinctions in different tissues. This result implied that Sox genes perhaps participated in development of testis and heart of Eriocheir sinensis.Difference of Sox gene expression between testis and ovary indicated that Sox gene took part in sex determination.This research provided molecular data for the sex-determining mechanism of Eriocheir sinensis and expression pattern of Sox gene...
