Laccase belongs to the environmental friendly enzymes,but laccase has the disadvantages of low environmental tolerance and easy inactivation,which has not been widely used.Therefore,this study put forward to use the effect of electric field to improve the enzyme activity.Laccase from Pleurotus ost reatus was regarded as the research object to study the characteristics of laccase,the effect of direct-current electric field on laccase,and the degradation effect and mechanism of phenol by the direct-current electric field on laccase.The main conclusions are as follows:1.The molecular weight of laccase was 40 KDa,and the isoelectric point was 4.51.and laccase could belong to copper glycoprotein with acidic protein features.The optimum pH and temperature for laccase activity were 3.0 and 40°C,but laccase had poor stability in the acid environment?pH=3.0?.Different solution conditions?kinds of ions,surfactants,organic solvents,etc.?could affect the properties of laccase.Compared with nitrogen heterocyclic chemicals and polycyclic aromatic hydrocarbons,the degradation of phenolic compound by laccase was better.2.The activity of laccase was enhanced in the anodic region,but at the same time was inhibited at the cathode,and electric field enhanced laccase stability in comparison with laccase with no applied electric field.The electric charge characteristics of laccase were changed when exposed to electric field,and some laccases molecules moved to the anode,which produced a slight migration phenomenon.The effect of electric field on laccase had better electricity and biology effects under the condition of suitable temperature?20-40°C?,pure water environment,low voltage,uniform electric field.The complex solution state under the electric field?buffer system,C uSO 4,Tween-80,methanol solvent?changed the effect of electric field on laccase.3.The degradation process of phenol in the elec tric field and the properties of laccase activity,the characteristics of material migration were consistent with one another.The degradation of phenol by laccase was dominant role,and the degradation by electric field was supplementary role.Phenol had better removal efficiency under the condition of suitable temperature?20-40°C?,pure water environment,uniform electric field.The complex solution state under the electric field also affected laccase on phenol oxidation process,and in the pH=3.0 buffer system,CuSO4 solution system,Tween-80 solution system,phenol had changed the degradation mechanism.
