The essence of this study on the relationship between transportation infrastructure and manufacturing productivity is to evaluate the economic benefit of the transportation infrastructure investment comprehensively, and then provide relevant suggestions for government making some policies on the transportation infrastructure investment.Previous scholars mainly focused on the benefits of transportation infrastructure and the relationship between transportation infrastructure and economic development. They mostly ignored the spatial spillover effects of transportation infrastructure and rarely analyzed the specific mechanism. And also they adopt the provincial data. This paper aims to reveal the spatial spillover effects of transportation infrastructure to the manufacturing productivity in the Yangtze River Delta urban agglomerations.Firstly, we analyze the mechanism on the spatial spillover effects of transportation infrastructure: by reducing manufacturing enterprise inventory cost, accelerating the regional market integration and promoting the manufacturing industry agglomeration.Secondly, through using the DEA-Malmquist Index, we get the total factor productivity and technical efficiency and technical progress of manufacturing industry in 2002-2013.Thirdly, by using Exploratory Spatial Analysis, we get the conclusion that there is some spatial correlation between the manufacturing productivities. The global Moran’s I index, the local Moran scatter plot and LISA agglomeration all prove it. Finally, by controlling some economic factors, such as FDI,R&D, we make focus on the spatial spillover effects in the manufacturing productivity of highway. The results show that if we add the interaction between the transportation infrastructure and scale of the neighbor city, the output elasticity of local transportation infrastructure will rise from 0.765 to 1.146, indicating that the neighbor city’s scale will promote local economy through transportation infrastructure. The spatial spillover effect from transportation infrastructure is largely positive, because it reduces the output elasticity of local transportation infrastructure from 0.765 to 0.628. The adjacent transportation infrastructure elasticity of manufacturing productivity is 0.120.
