The invention of the microscope makes it possible for people to observe the microscopic structure of the cells. Therefore, it makes people finally find out that the nature of the organ disease is the cell lesion. Meanwhile, the pathology changes with a qualitative leap. The pathology has gone through the levels of naked-eyes, optical microscope and electronic microscope. With the rapid development of computer technology, pathology entered the digital age in 1990 s. The emergence of digital pathology helps doctors solve some shortcomings, such as observing the microscope alone for a long time and being inconvenient for them to communicate with each other. It should be pointed out that the development of digital pathology is due to the emergence of digital slide, which is obtained by the digital slide scanner system. Different kinds of scanning techniques are emerging one after another.Subsequently, there are many kinds of image acquisition techniques, new lighting systems and image processing techniques.At present, the research regarding digital slide scanners has entered the period of rapid development. Accordingly, the related products are updating quickly and the technical requirements continuously enhance. The accuracy and the speed of digital imaging are good examples. In this paper, a new method is proposed for the above problems. First of all, macro camera is set up to obtain the navigation map of the slide, which is the organization range on the slide. Secondly, the high-power lenses are utilized to get organization details of the samples by meticulously scanning the organization part on the navigation map.This paper focuses on the dark-field illumination techniques in order to improve the contrast of navigation map on the digital slide. When scanning the fluorescence slide, it is of significance to obtain the organization range of the slide as the navigation map in advance.Under bright-field illumination, the fluorescence is very weak. Therefore, it is hard to distinguish between the dust on the cover glass and the organization of the slides, resulting in inaccurate identification of the map. Furthermore, for different kinds of pathological slides,such as the ones with different color depths of sample organization and different tissue thicknesses, various illumination approaches should be applied in order to get a more accurate navigation map. This paper aims to design a reasonable dark-filed illumination system so that the efficiency of distinguishing the dust and the sample organization on the fluorescence slide could be improved. Meanwhile, the factors affecting the imaging contrast will be obtained in the experience. Finally, reasonable solutions will be come up with. Additionally, this paper focuses on single-chip and slide clipping scanners and four different types of dark-field illumination are proposed, which are total reflection illumination, LED back-illumination,diffuse reflection illumination and oblique illumination. Moreover, this paper will contain an analysis on principles and characteristics of all kinds of illumination. According to the existing equipment of digital slide scanners, respective reasonable ways to achieve dark-field illumination will be come up with. A conclusion will be made after the analysis to find out the easier ways to get higher-contrast and accurate navigation map depending on different slides among those kinds of dark-field illumination. By comparing with the navigation map scanned from 2× objective lenses of the VM1000 digital slide scanner, the image contrast is slightly improved or equal when four different kinds of dark-field illumination techniques proposed in this paper are applied to the respective applicable pathological slides. However, the scanning speed of the illumination and scanning modes mentioned in this paper has improved more than 100 times.
