Among cancer patients, the mortality rate of leukemia has ranked in the top. And it is one of the major human life killers. In recent years, it has focused wide attention on purification technology in vitro of autologous peripheral blood with microwave radiation in the treatment of leukemia people for its convenience, safety and low prices. However, it has a high recurrence rate. The main reason is that the temperature and time of microwave radiation can’t be controlled to ensure killing the leukemia cells effectively and protecting the normal cells at a minimum degree of injury. This paper develops a in vitro purification system of autologous peripheral blood stem cells with microwave radiation based on cell pattern recognition and automatic control theory. The paper’s main work shows as follows:First, the overall system program has been completed. This system is mainly composed of two parts:the software system and the hardware system. With the basis of comprehensive study on the characteristics of the related image processing algorithms, the image processing algorithms for system software development were determined.Second, according to the obvious difference of brightness and morphological between leukemia cells’ active state and death status, a new leukemia cell death recognition method was studied based on the luminance difference between cell membrane and background. The method was validated with the dead cells’ consensus form, and the results showed that this method was feasible. By using it, the cells’death status in microwave radiation can be identified in real time. The recognition accuracy rate is about87%.Third, this paper designs the hardware circuit about in vitro purification system of autologous peripheral blood stem cells with microwave radiation, achieving two automatic control functions:①When the real-time temperature in the purification materials is higher than40℃, the hardware circuit will stop the microwave radiation automatically; when its temperature is lower than38℃, the hardware circuit will re-open the microwave radiation.(2)Under the condition that the purification’s temperature varies in the range of38~40℃, if the cell death rate in real time exceeds the predetermined threshold, the hardware circuit will stop the microwave radiation automatically. STC12C5A60S2MCU is the control core in the circuit, and it mainly consists of eight parts:temperature acquisition module, serial communication module, switch control module, etc.Last, the system completed the hardware and software integration and test. The results showed that the in vitro purification system of autologous peripheral blood stem cells with microwave radiation based on pattern recognition is stable and has full functionality. It has reached the expected goal.
