Objective:this research is to study the nutritional obesity model built by using the high-calorie diet to feed SD rats, and experimental intervention in obese rats with swimming training and food control. On the one hand, observe the influence on the two obesity indexes, weight and the lee’s index, made by swimming training on different time combined with food control. Explore the results of obesity made by swimming training combined with food control; on the other hand, by using a radioimmunoassay and RT-PCR experimental methods, explore changing rules of the central and peripheral neuropeptide Y in obese rats immediately after different training time and in recovery time to reveal the impact of swimming training and food control on the hypothalamic control of food intake regulation in obese rats, for further study the regulation of food intake in patients with obesity mechanism to provide a theoretical reference.Object and methods:In this study,220 clean SD female rats were randomly divided into normal control group fed with normal diet and obesity model group fed with high-calorie diet. After 6-8 weeks, build nutrition obese rats, and record daily food intake of rats in each group everyday, recorded each subject’s body weight, body length and Lee’s index once a week. After successful modeling obesity model, proceed the intervention experiment by adopting the method of swimming training and food control, generally it can be divided into three groups according to the intervention time:two weeks, four weeks and six weeks, each group are further divided into the normal control group, the obesity control group, immediately observation exercise group and after 3 hours observation exercise group. The normal control group was not limited in normal diet and do not exercise, the obesity control group does not limit high-calorie diet and no exercise. In the experimental group, obese rats were trained for two weeks, four weeks and six weeks,60min/day, 6 days swimming/week combined with food control which provide normal diet. After the combination of swimming training with control eating plan, Anatomy drawn blood, brain and hypothalamus. Respectively, using radioimmunoassay and RT-PCR experiments were detected hypothalamic neuropeptide Y mRNA expression level of the lower periphery of plasma neuropeptide Y in the system and the central nervous system.Results:1.200 SD rats by feeding high-calorie diet for 8 weeks to build successfully 62 high-calorie diet-induced obese rats.2. With the increase of the intervention time of the combination of experimental swimming with food control, weight of the obese rats and Lee’s index gradually reduced. Until in the end of the 6th week, it dropped to normal levels in the control group.3. Four weeks of swimming combined food control can reduce plasma neuropeptide Y in obese rats immediately after exercise when hypothalamic neuropeptide Y mRNA expression levels, and 3 hours after training began to rise to the level of obesity control group;4.6 weeks of swimming combined food control intervention immediately after exercise, and 3 hours after training of neuropeptide Y in plasma in obese rats, hypothalamic neuropeptide Y mRNA expression levels did not have a significantly different affect, but the two groups were significantly lower than those in obese control group;Conclusion:1. Feeding combining a certain time of swimming training can effectively reduce the fat rat body weight, Lee’s index,and in comparison with the experimental intervention of different weeks,4 weeks effect reducing weight is better than 2 weeks,6 weeks experimental intervention so much that the obese rats reached the effect reducing weight.2. Feeding combining the swimming training can reduce the content of obese rats plasma neuropeptide Y and the neuropeptide Y mRNA gene expression levels, which affects the body’s eating behavior, ultimately lead to weight loss in obese rats, and the experimental intervention, the longer the better the effect reducing weight.3. This experiment can not reveal the way other sports combined with food control to impact of control changes of NPY obesity, the theory of weight loss attributed by the combination of the movement and food control needs in-depth study to learn more about the influence of obesity on the body made by sports combined with control food.
