Objective: To explore effects of different exercise intensity combination with CLA on blood lipid,the expression of PPARγ m RNA gene,protein expression in visceral adipose tissue, plasma concentration of PPARγ in adolescent obese rats.Methods:Obese rats were modeled with high fat feeding, 64 obese rats were selected and were randomly divided into 8 groups, 8 rats each group,Including control group, resting+CLA group,low intensity exercise group,low intensity+CLA group,strength intensity exercise group, strength intensity+CLA group, high intensity exercise group, high intensity+CLA group,Each exercise group is trained by treadmill training for 8 weeks. High,strength,low,exercise treatment were performed on animal treadmill with running speed of 15-18m/min, 21-25 m/min, 28-32 m/min, 60min/Every day, 5 times / week,low intensity+CLA group, strength intensity+CLA group,high intensity+CLA group with CLA(1.6g/kg)by gavage, 1 times /day, 5 times / week,for 8 weeks. blood and adipose tissue are collected After 8 weeks, blood lipid is measured, PPARγ m RNA gene expression in adipose tissue was tested using q RT-PCR, PPARγ protein expression in adipose tissue by immunohistochemistry,concentration of PPARγ in plasma by ELISA.Results: Different exercise intensity and CLA supplementation on adolescent obesity rats after 8 weeks intervention, Blood lipid, the expression of PPARγ m RNA gene, protein expression in visceral adipose tissue, plasma concentration of PPARγresults show:1 Blood lipid(1) 8 weeks of different intensity exercise intervention,TC concentration of high,strength intensity group is lowed than control group(P<0.05),TG concentration of low, high,strength intensity group is lowed than control group(P<0.05), LDL-c of high,strength intensity group is higher than control group(P<0.05).( 2) 8 weeks of different intensity exercise and CLA intervention, TC concentration of strength intensity, resting+CLA group is lowed than control group( P<0.05),TG concentration of strength intensity group, strength intensity+CLAgroup is lowed than control group, resting+CLA group(P<0.01), LDL-c of strength intensity group, strength intensity+CLA group is higher than control group,resting+CLA group(P<0.05), there is no difference in groups in HDL-c(P>0.05).(3)8 weeks of different intensity exercise Combination with CLA intervention, TC concentration of high intensity+CLA group, resting+CLA group is lowed than control group(P<0.05),TG concentration of high intensity+CLA group, strength intensity+CLA group, low intensity+CLA group is lowed than control group, resting+CLA group(P<0.01, P<0.05), LDL-c of high intensity+CLA group, strength intensity +CLA group, low intensity+CLA group is higher than control group, resting+CLA group(P<0.01, P<0.05), The above indices(except TC)control group and+CLA group had no difference(P>0.05).2 The expression of PPARγ m RNA gene in visceral adipose tissue( 1) 8 weeks of different intensity exercise intervention, the expression of PPARγ m RNA gene in visceral adipose tissue in high,strength low intensity group is higher than control group(P<0.05), strength low intensity group is higher than high intensity group(P<0.05).(2)8 weeks of different intensity exercise and CLA intervention, the expression of PPARγ m RNA gene in visceral adipose tissue in strength intensity group, strength intensity+CLA group is higher than control group, resting+CLA group(P<0.01), the expression of PPARγ m RNA gene in visceral adipose tissue in resting+CLA group is higher than control group, but there is no different(P>0.05).(3)8 weeks of different intensity exercise Combination with CLA intervention,the expression of PPARγ m RNA gene in visceral adipose tissue in high intensity +CLA group, strength intensity+CLA group, low intensity+CLA group is higher than control group, resting+CLA group(P<0.01), high intensity+CLA group is higher than strength intensity+CLA group, low intensity+CLA group(P<0.05).3 Plasma concentration of PPARγ(1)8 weeks of different resting+CLA group(P<0.01), plasma concentration of PPARγ in resting+CLA group is higher than control group, but there is no different(P>0.05).(3)8 weeks of different intensity exercise Combination with CLA intervention,plasma concentration of PPARγ in high intensity+CLA group, strength intensity+CLA group, low intensity+CLA group is higher than control group, resting+CLA group(P<0.01),high intensity+CLA group is higher than strength intensity+CLA group, low intensity+CLA group(P<0.05).4 The expression of PPARγ protein in visceral adipose tissue:8 weeks of different intensity exercise reduce the concentration of TG and TC,improve the expression of PPARγ m RNA gene in visceral adipose, PPARγ protein expression, plasma concentration of PPARγ, strength low intensity group is higher than high intensity group.(2)through 8 weeks Different exercise intensity Combined with CLA intervention of adolescent obesity rats can reduce the concentration of TG,improve PPARγ m RNA gene expression and protein expression in visceral adipose tissue of increased significantly, plasma concentrations of PPARγ increased significantly, strength low intensity group is higher than high intensity group.(3)Simple exercise is better than the simple complement CLA for adolescent obese for concentration of TG and TC, the expression of PPARγ m RNA and protein in adipose tissue and plasma PPARγ concentration, and Other related indicators, There is no difference between sports with CLA and pure exercise.
