Objective: With an animal experiment, by testing rats lee’s index and the changes of plasma FFA, IL-6 levels, comparison and analysis the liver FFA, IL-10 content and Chemerin/CMKLR1-m RNA、KCs expression and invasion, to explore the effect of high-fat diet on lipotoxicity, as well as the protective effects of treadmill exercise for 6 weeks combined with Chlorella on high- fat diet rat liver cells.Methods: Got 46 clean,8-week-old male SD rats,weight 200g± 20 g, were randomly divided into five groups after 1 week feeding adaptation : control group(N = 10, C), high-fat diet group(N = 10, H), fat diet exercise group(N = 10, HE), high-fat dietary supplement Chlorella group(N = 8, HL), high-fat dietary supplement Chlorella and exercise group(N = 8, HEL). Group C normal diet routine feeding, H group, HE group, HL group, HEL fat diet feeding group. HE 、HEL group with aerobic exercise 0 slope are used treadmill exercise. Running speed : the first week of 15m/min, the second week of 20m/min, after that an increase 1m/min week. Times 50min/times. Sports Time :21:00-24:00,1 time/day, 6 days/week, and last six weeks. Group HL and HEL administered Chlorella(20mg/kg body weight), the other group fed the same amount of volume of saline water, administered 30 minutes before the time of exercise. The first 6 weeks, the groups were fasted for 12 hours, the rat tail vein blood fasting glucose, blood plasma was separated after the apex 30 min and the supernatant was centrifuged at 3000 rpm placed in vials stored at-80 ℃tested quickly removal of the liver, were placed in liquid nitrogen and stored in formalin for later use. ELISA method to measure plasma cytokines IL-6, insulin(Fins),FFA and liver homogenate IL-10, FFA level; using PCR technology to detection the level of CMKLR1-m RNA expression; Western blot detection the expression levels of Chemerin protein; HE staining of liver tissue infiltration observed KCs. Test using SPSS17.0 software processing the data obtained.Results: 1. Compared with group C, group H of rats Lee’ s index, plasma FFA IL-6,FFA, IL-10 levels of liver was significantly higher(P<0.05,P<0.01), chemerin/ CMKLR-m RNA expression was up-regulated(P<0.05).2. HE staining of liver tissues showed diffuse, vesicle like steatosis and Kupfer cell infiltration. HOMA-IR increased, but showed no significant difference(p>0.05).3. Group HE,HL,HEL rat plasma FFA,IL-6 levels and HOMA-IR were lower than group H(p <0.01).Effects of aerobic exercise,Chlorella supplements on plasma FFA and IL-6 showed significant interaction.Meaning that the same movement condition,lee added Chlorella can make the rats plasma FFA,IL-6 levels decreased(p <0.01).But Effects of aerobic exercise, supplement Chlorella on plasma HOMA-IR no significant interaction(p>0.05).4. Group HE,HL,HEL rat liver FFA levels and chemerin / CMKLR-m RNA were lower than group H(p <0.01) 、 IL-10 increased, but showed no significant difference(p>0.05). Effects of aerobic exercise, supplement Chlorella on FFA and IL-10 levels no significant interaction(p>0.05). Effects of aerobic exercise, supplement Chlorella on liver chemerin / CMKLR-m RNA expression showed significant difference(P<0.05), Meanwhile supplement Chlorella, effects of aerobic exercise on chemerin / CMKLR-m RNA significant interaction(p<0.05).5. Group HE, HL, HEL rat liver tissue HE staining, vesicle like steatosis degree reduction.Conclusion: 1. 6 weeks of high fat diet rats were lee’s index increased significantly, The increased of plasma FFA and HOMA-IR levels in different degrees, the body showed lipotoxicity,inflammation state and IR.2. 6 weeks of high fat diet rats, due to liver FFA, IL-10 level increase, fatty degeneration of hepatocytes, infiltration of KCs, Chemerin/CMKLR1-m RNA expression level increased, induced liver lipotoxicity and inflammation in rat liver.3. 6 weeks aerobic exercise or supplement Chlorella can reduce weight.reduce body and liver lipotoxicity, relieve body inflammatory state improve insulin resistance, has the function of protecting liver. Aerobic exercise combined added Chlorella on liver lipotoxicity protection than simple aerobics or added Chlorella more effective.
