Study On Xiaoliu Granule For Treating Hysteromyoma And For Influence Of Hysteromyoma In Rats

Objective:To observe the clinical effect of Xiaoliu granule to treat hysteromyoma and investigate the influence of Xiaoliu granule on the experimental hysteromyoma in rats and explore the mechanisms of Xiaoliu granule treating hysteromyoma.Methods:Clinical research:41 cases of hysteromyoma were treated by Xiaoliu granule.During the study,we observed and recorded the Patient’s changes of clinical symptoms and signs.At the beginning and the end of treatment,we monitored the changes of the uterus and myoma with B-us.Then we compared the changes of the symptoms and signs、the uterus and the myoma.Experimental investigation: Hysteromyoma rats were injected with estrogen and progesterone together,treated with Xiaoliu granule and Guizhifuling capsule.At the end of experiment,we calculated the uterus index and observed the changes of the smooth muscle under microscope.Expressions of estrogen receptor(ER),progesterone receptor(PR)and bcl-2 protein were examined by immunohistochemistry.Result:Clinical research:Xiaoliu granule can effectively treat the clinical curative, the total effective rate is 80.5%.Experimental investigation:1 The gross appearance and observations under microscope showed the pathologicalproperty of the uterus. Compared with the controls,weight of the uterus,uterus index,rats were increased significantly(P＜0.05),the myometrium of treated rats were thickened and the smoothmuscle cells enlarged(P＜0.001).And more connective materials constructed the extracellular matrix.It resulted that we successfully built up the uterine leiomyomas models in rats by injecting estrogen and progesterone;2 Xiaoliu granule preparation presented the effects to inhibit the myometrium proliferation,to lighten the uterine weight,to decrease the uterus index(P＜0.05),microscopically,Xiaoliu granule was effective,to reverse the proliferative abnormalities of the focal smooth muscle(P＜0.05);3.The hyper-status of ER and PR in the focal tissue were important factors to accelerate the development of leiomyoma,one mechanism of Xiaoliu granule preparationcuring the uterine leiomyomas was to inhibit the expressions of ER and PR(P＜0.05),and to attenuatethe proliferation;4 It resulted in local uterine smooth muscle abnormal apoptosis that the reinforcement of the expression of the inhibiting apoptosis gene Bcl-2(P＜0.05),which was anthor factors to accelerate the development of leiomyoma,anther mechanism of Xiaoliu granule preparationcuring the uterine leiomyomas was to rebalance the physiological apoptosis of leiomyoma cells by down-regulatingthe expression of bcl-2 protein.Conelusion.In summary,Xiaoliu granule have therapeutic function to hysteromyoma; According to the experimental investigation,the hyper- status of estrogen and progesterone played an important role in improving the process of fibrosis and the inhibition of programed cell death enhanced the development of uterine leiomyomas. Xiaoliu granule preparation displayed remarkably inhibitory effects on development of uterine leiomyomas,its mechanism concern with regulate the concentration of estrogen and progesterone and resume apoptosis of leiomyomas cell.
