Angelica keiskei containing a variety of chalcone, coumarin, and plant nutrition,which has many pharmacological effects, such as unique hypolipidemic, anticancer, protecting liver and nerve function. At present domestic and international research of Angelica keiskei mainly concentrated in separation of chalcone ketones material qualitative and mechanism research, but lack research of differences in the distribution of the effective ingredients in different parts and antioxidant of Angelica keiskei. This study using Angelica keiskei as raw materials, respectively of Angelica keiskei root, stem and leaf of basic chemical composition and separation and purification process of ethanol extracts, and related research antioxidant activity of purified ethanol extract. The main conclusions are as follows:1. Determination content of Angelica keiskei different parts main chemical composition. Found composition distribution differs in different parts of Angelica keiskei. The basic chemical components in the leaves of Angelica keiskei were higher, the content of stem was the second, and the root was the lowest, and the content distribution of water-soluble flavone was: Angelica keiskei leaf > stem > root. Overall, Angelica keiskei leaf has highest values.2. HPLC-PDA was used to determine the content of 4-hydroxyderricin and xanthoangelol in root, stem and leaf of Angelica keiskei. 4-hydroxyderricin content distributed in Angelica keiskei was as follows: stem(149.16μg/g) > root(122.42μg/g) > leaf(1.24μg/g); xanthoangelol content distribution in Angelica keiskei was as follows: root(144.28μg/g)> stem(75.17 μg/g) > leaf(1.21 μg/g).3. Through the analysis of characteristics of static, dynamic adsorption and kinetic curve, determing use the HP-20 macroporous adsorption resin to purified the Angelica keiskei ethanol extract. The flavone content of ethanol extract of root, stem and leaf of Angelica keiskei were 82.33, 19.33, 7.35 mg/g, the flavone content of purified ethanol extract of root, stem and leaf of Angelica keiskei were 180.48, 83.21, 57.67 mg/g. By flavone chromogenic qualitative reaction, speculating ethanol extracts of different parts of Angelica keiskei may contains 5-OH, or 2-OH group chalcone, 3-OH or o-dihydroxybenzene, 5-OH, 4-ketone group, phenolic hydroxyl or flavonols.4. The total antioxidant, scavenging free radicals and anti-lipid oxidation capacity of purified ethanol extracts of different parts of Angelica keiskei have been improved by DPPH, FRAP and linoleic acid peroxidation inhibition test. Concentration was 1.09 mg/ml ethanol extract of Angelica keiskei root, DPPH free radical clearance rate was 53.11%, the total antioxidant capacity is as same as 0.411mmol/L FeSO4 solution, and linoleic acid peroxidation inhibition rate was 10.56%; concentration was 107.5 mg/ml ethanol extract of Angelica keiskei stem, DPPH free radical scavenging rate was 78.86%, the total antioxidant capacity is as same as 0.703 mmol/L FeSO4 solution, and linoleic acid peroxidation inhibition rate was 15.56%; concentration was 1.05 mg/ml ethanol extract of Angelica keiskei leaf, DPPH radical scavenging rate was 88.86%, the total antioxidant capacity is as same as 1.071 mmol/L FeSO4 solution, and linoleic acid peroxidation inhibition rate was 16.10%.
