This dissertation has two parts. The first part is the extraction and purification of alpinetin and cardamomin from Alpinia katsumadai Hayata and the preliminary research on DPPH free radicals scavenging. The second part is the optimized extraction of the total flavonoids from Angelica keiskei koidzmi and the preliminary composition analysis and DPPH free redical scavenging activity research.Alpinetin and cardamomin are the major effective ingredients, which exist in Alpinia katsumadai Hayata, belonging to flavonoids compounds.They have strong activity in inhibiting platelet aggregation and tumor formation, anti-inflammatory, antibacterial as well. They are in a low toxicity and have good safety.Alpinetin and cardamomin were extracted from Alpinia katsumadai Hayata by solvent extraction method, and their contents were analysed by high performance liquid chromatography (HPLC).Using heating reflux method and microwave extraction, two optimized extraction processes were obtained, respectively.The results show that two methods have good extraction effect. Using Alpinia katsumadai Hayata powder as raw material, single factor experiments and orthogonal experiments were tested.The best technological conditions of heating reflux method were as following:100%ethanol,20times solid-liquid ratio, extracted2hours and three times. The best technological conditions of microwave extraction were as following:100%ethanol,12times solid-liquid ratio, power800W, extracted12min and two times. Microwave extraction can greatly reduce the extraction time, the extraction ratio of alpinetin and cardamomin are8.73mg/g and0.99mg/g. After extracted by ethyl acetate, the contents of alpinetin and cardamomin in crude extract are10.15%and1.27%.High-speed counter-current chromatography (HSCCC) was used to separate and isolate of alpinetin and cardamomin from Alpinia katsumadai Hayata. The two-phase solvent system was composed of n-hexane-ethyl acetate-ethanol-water (5:6:5:5, v/v).The optimum separating conditions were25℃of column temperature,2.0mL/min of the flow rate,850rpm of revolution speed and detected at280nm.14.47 mg of alpinetin and1.72mg of cardamomin were obtained from158.4mg of the crude extract. The recovery and purity of alpinetin was90.01%and99.01%, respectively. The recovery and purity of cardamomin was85.70%and97.71%, respectively.Their structures were confirmed by melting points, IR,1H NMR and13C NMR. The puried alpinetin and cardamomin is tested for DPPH free radicals scavenging activities. The results show that alpinetin had strong DPPH free radicals scavenging activities.Angelica keiskei koidzm, which is the primary parsley family herbaceous perennial plant, is rich in unique elements, chalcone and coumarin. Chalcone has a role in anticancer virtue, antibacterial, allergy, AIDS, inhibiting the gastric acid fight ulcer, preventing blood clots and diabetes functions.Three solvent extraction methods were used to extract the total flavonoids from Angelica keiskei koidzm. The optimum extraction technology of the heating reflux method, microwave extraction and ultrasonic extraction were tested. Among them, heating reflux method is the best, but is extraction time is the longest. Using UV-Vis spectrophotometer, the total flavonoids contents were tested by the NaNO2-Al(NO3)3-NaOH development process and AlCl3development process. Both of them are stable and reliable. The flavonoids contents tested by the NaNO2-Al (NO3)3-NaOH development process were higher than that by AlCl3development process.Infrared spectra showed that the extracts are flavonoids compounds, and color reaction showed that flavonoids ingredients may be chalcone, two hydrogen flavonoids compounds. The total flavonoids of Angelica Keiskei koidzm was tested for DPPH free radicalss scavenging activities. The results show that the Angelica keiskei koidzm extracts have strong DPPH free radicals scavenging activities.
