Objective To study the effects of propofol on the proliferation,apoptosis and secretion of NO in mouse glomerular mesangial cells(GMCs) stimulated by lipopolysac charide.Methods The mouse glomerular mesangial cells(GMCs,SV40MES13) were cultured to 3-5 generations and collected routinely.Then GMCs were planted in a 96-well culture board and two 48-well culture boards uniformly after the concentration be adjusted.The cells were divided into eight groups,including blank control group,propofol(5,10,20ug/ml) groups,LPS(1ug/ml) group and LPS(10ug/ml)+propofol(5,10,20ug/ml) groups.Each group was set 6 wells.The corresponding stimulant was added to cells respectively in accordance with above grouping.Different doses of propofol was added in cells after the addition of LPS 1 hour in LPS(10ug/ml)+propofol(5,10,20ug/ml) groups.Finally the GMCs were cultured in cell incubator for 48 hours.After the GMCs cultured 48 hours,the cell growth was observed under the inverted microscope and photographed.Then the cell proliferation was detected by MTT.The cell culture medium was collected and the content of NO was measured by ELISA.The cell apoptosis was detected and analyzed by flow cytometry.Results 1 The cell growth was observed under the inverted microscope and the number of cells in LPS group was more than the blank control group and propofo l alone groups obviously.Moreover, the cells changes from irregular to circular.The number of cells in LPS +propofol(10,20ug/ml) groups was less than LPS group.However,the chang of LPS+propofol 5ug/ml group was inconspicuous. 2 There was no noticeable difference between the propofol alone groups and the blank control group in cell proliferation,apoptosis rate and NO secretion(P>0.05).Compared with the blank control group,the cell proliferation and NO secretion of LPS group was significantly increased,but the apoptosis rate decreased(P<0.05).However,the proliferation degree and NO secretion was weakened and the apoptosis rate was increased in LPS+propofol(10,20ug/ml) groups compared with the LPS group(P<0.05). Mean while, this results is more obvious in LPS+ propofol(20ug/ml) group(P<0.05). B ut there was no significant change between LPS group and LPS+propofol(5ug/ml)group(P>0.05).Conclusions 1. Propofol in low,medium or high concentrations(5,10,20ug/ml)has no effect on the proliferation,apoptosis and secretion of NO in GMCs. 2. Propofol in medium or high concentrations(10,20ug/ml) can inhibit the proliferation, NO rele ase and promote the apoptosis of GMCs stimulated by LPS and play a part in kidney protection. Meanwhile, this results is more obvious in LPS+propofol(20ug/ml)group(P<0.05). But propofol in low concentration(5μg/ml) does not have such effect.
