Objective To detect the effect and underlying mechanism of Parthenolide(PN) on intimal hyperplasia in iliacartery of rabbit after balloon injury.Methods The inflammatory model of intimal hyperplasia was established by a combination of balloon injury and high-fat diet. After 1 week of high-fat feeding, 30 male white rabbits(2.0-2.3) kg were randomly divided into 6 groups: A:Sham+NS: rabbits got 0.9% normal saline(NS) after sham operation(n=5); B:Sham+DMSO: rabbits got 4%DMSO after sham operation; C:balloon injury+NS: rabbits got 0.9%NS after balloon injury(n=5); D:balloon injury+DMSO: rabbits got 4%DMSO after balloon injury(n=5); E:balloon injury+PN low: rabbits got PN at 1mg/kg after balloon injury(n=5); F:balloon injury+PN high:rabbits got PN at 2 mg/kg after balloon injury(n=5). Drugs were intraperitoneal injected once per day until sacrifice. After high-fat diet for four weeks, 30 rabbits were all seleceted for this study. The intima-media thickness, the expression of NF-κB, IL-6, Caspase-1, IL-1β, and the levels of IL-8, TNF-α, TC, TG, LDL, HDL in serum were measured.Results The results of Comparison between group A vs group B and group C vs group D showed that there was no statistical significance(P>0.05). Compared with group B(Control gourp), the thickness of intima, the expression of NF-κB, IL-6, Caspase-1, IL-1β, IL-8 and TNF-α in group D(Model gourp) were significantly increased(P<0.05). The amount of NF-κB, IL-6, Caspase-1, IL-1β, IL-8 and TNF-α was significantly decreased in F group(High-dose group) compared with D group(P<0.05). The level of lipid was not statistically significant in different groupsConclusion Neointimal hyperplasia is suppressed by PN after balloon injury, the potential mechanism maybe associate with its anti-inflammatory role in inhibiting the activation of NF-κB and Caspase-1 and down-regulating the expression of inflammatory factors.
