ObjectiveTo investigate the effects of transplanting adipose tissue-derived mesenchymal stem cells on intimal hyperplasia after carotid balloon injury and the possible mechanism.Method45 male Sprague Dawley rats were randomly divided into three groups:sham group( n=15),experimental group(balloon injured, n=15),ADMSC group(balloon injury and treatment with ADMSC, n=15).the balloon catheter was inserted into the left carotid to produce intimal injury,neck midline incision alone was performed in sham group.After 1,2,4 weeks,rats were sacrificed and the injured carotid artery and normal carotid artery were used for HE-stained pathological study.The m RNA expressions of e NOS were determined respectively by RT-PCR. and the protein expressions of p21、p27 and PCNA were determined respectively by Western Blotting. Changes of the expression levels of NO were detected by enzyme-linked immunosorbent assay(ELISA).Results1. Enhanced green fluorescent protein positive cells were localized to the traumatized arterial segment within 1 week after transplantation.2.Compared with sham group, the W/L ratios of experimental group and ADMSC group was increased obviously,with a significant difference in statistics(P<0.05).In addition,the W/L ratios of ADMSC group were lower than experimental group.3.The protein expression of p27、p21 significantly increased and reached a peak at four weeks after balloon injury(P<0.05),compared with sham group.Otherwise,the protein expression of PCNA significantly increased and reached a peak at 1 weeks after balloon injury(P<0.05),compared with sham group.4.In comparation with injury group, ADMSC treatment significantly increased theprotein expression of p27、p21(P<0.05).And after one week treatment,the expression of PCNA significantly decreased(P<0.05),compared with experimental group. 5.The m RNA expressions of e NOS were significantly than the experimental group after four weeks treatment.(P<0.05).6.the level of NO in plasma were significantly increased after four weeks treatment(P<0.05),compared with experimental group.ConclusionADMSC could localized to the traumatized arterial segment and inhibit the process of intimal hyperplasia by upregulating the expression of protein p21、p27 and e NOS m RNA,as well as the level of NO in plasma.
