Background: TRIM44(Tripartite motif-44)belongs to TRIM family, which plays an important role during neuronal differentiation and maturation. Studies have shown that stomach esophageal cancer and breast cancer overexpressing TRIM44 are more aggressive. Meanwhile, TRIM44 overexpresses in mouse brain hippocampus site,which suggest TRIM44 may also play an important role in glioblastoma.Objective: To estimate the expressing of TRIM44 in glioblastoma cell lines and observe the influence of TRIM44 silencing on tumor cell function. We try to probe the related molecular mechanisms in order to decide whether TRIM44 can be the therapy target and prognostic markers during the treatment of glioblastoma.Methods: Evaluate the expressing of TRIM44 in glioblastoma cell line by Real-time PCR. Construct TRIM44-shRNA lentivirus vectors which can knock down TRIM44.Then detect the changes of glioblastoma cells’ function on cell proliferation, colony formation, migration, and apoptosis. Using Luciferase Reporter Assay System, we try to preliminarily explore the role of TRIM44 in the development of glioblastoma and the potential mechanisms.Resμlts: TRIM44 expression in human glioblastoma cell lines is higher than that in normal cell lines. The glioblastoma cells’ proliferation, colony formation and migration ability all reduced obviously after silencing TRIM44 by shRNA lentivirus infection. FCM showed that apoptosis rate increased after TRIM44-sh RNA lentivirus vector infection. TCGA database shows that among 160 cases of glioblastoma, TRIM44 gene of 98 cases fuse accounting for 61.25 percent. Dual luciferase reporter gene experiments showed the luciferase activity of control group(plenti-utr-luc-blank) is about 2.5 times that of the experimental group(TRIM44 3’UTR Lenti-reporter-Luc).Conclusion: TRIM44 plays oncogene role in glioblastoma and silencing TRIM44 can impaire the proliferation, cell migration, colony formation of glioblastoma cells. We preliminarily come to the conclusion that TRIM44 can be the therapy target and prognostic markers during treatment of glioblastoma. At the same time,detecting the role of TRIM44 can provide clues for individualized treatment of glioblastoma patients.
