Purpose In order to detect the expression of PHF2 in human breast cancer cells and tissues, the biological function of PHF2 in breast cancer; then analyse the correlation between the expression of PHF2 and the clinical parameters, prognosisMethods The qPCR detected the expression level of PHF2 mRNA in breast cancer, normal human mammary epithelial cell lines HBL100 and paired breast cancer tissues; Western blot examined the expression of PHF2 protein in breast cancer cell lines. Immunohistochemistry detected the expression of PHF2 protein in breast cancer tissues and adjacent breast cancer tissues, then analysed the relationship between the expression level of PHF2 and the clinical parameters, prognosis of breast cancer. The adenovirus vector system upregulated PHF2 gene, in breast cancer cell lines. The CCK8 kit, Colony formation investigated the ability of proliferation; Cell-cycle assay detected the distribution of cell cycle; the Wound-healing and Transwell assay probed the ability of migration and invasion of MDA-MB-231 and MCF-7. Moreover, xenograft tumor of null mice assay detect the effects of PHF2 on the proliferation of breast cancer.Results The PHF2 mRNA and protein were down-expressed in breast cancer cell lines(BT549, MDA-MB-231, MDA-MB-468, MCF-7, SK-BR-3, T47D) compared with normal human mammary epithelial cell HBL100(P<0.01). The PHF2 mRNA was down-regulated in 90%(27/30) breast cancer tissues compared with adjacent tissues(P<0.05), the PHF2 protein was down-regulated in 70%(35/50) breast cancer tissues and the low expression of PHF2 was significantly associated with the lymph node metastasis, negative ER statues and poor prognosis of breast cancer patients(P<0.05). Compared with the control group, the over-expressed PHF2 obviously inhibited the capability of proliferation, migration and invasion of breast cancer cells(P<0.01). In vivo, the ectopic expression of PHF2 significantly inhibited the proliferation of breast cancer(P<0.05).Conclusion The PHF2 frequently down-expression in breast cancer cells and breast cancer tissues; PHF2 could be as a marker of prognosis of breast cancer and may be a new target of breast cancer treatment.
