Objective:1.To investigate the CETP gene 10 polymorphism distribution in the control group and with dyslipidemia, serum lipids between relationship to abnormalities in Uygur and Kazak blood lipid.2.Whether there is a linkage disequilibrium phenomenon and the relationship between haplotype and Uygur, Kazak dyslipidemia the CETP gene 10 polymorphic loci revealed.Methods:1.Established in the survey based on the database(3049 of Kazakh, Uygur 5692), randomly selected from Uygur, Kazak blood lipid abnormality 367 cases and 345 cases as dyslipidemia group, and from 374 cases and 355 cases were randomly selected from the same population of blood lipid of the two nations normal individuals as the control group.2.Use the SNa Pshot SNP genotyping of CETP gene were detected in all subjects 10 polymorphism.3.Using SPSS 20 statistical software package was used to data processing, mainly using χ2 test, t test,single factor Logistic regression analysis, calculated odds ratios(OR) and 95% confidence intervals.Linkage disequilibrium and haplotype analysis were processed by SHEsis online software for processing.Results:1.Uyghur population CETP gene 10 SNP loci, rs12149545, rs3764261, rs1800775, rs711752 and rs708272 locus genotype and allele differences were statistically significant in dyslipidemia group and the control group(P <0.05). Wherein the dyslipidemia group A allele of rs12149545, T allele of rs3764261,T allele of rs711752 and A allele of rs708272 were lower than the control group(P <0.05). Dyslipidemia group rs1800775 C allele frequency the control group( χ2=7.475, P=0.006). rs289714, rs5882 and rs1801706 genotypes and allele frequencies did not differ in dyslipidemia group and the control group(P>0.05).2.Uyghur dyslipidemia group AA genotype of rs12149545 HDL-C levels higher than GG genotype.rs3764261 TT genotype of HDL-C levels higher than GG genotype. The CC genotype of rs1800775 low HDL-C levels in AA genotype, rs711752 AA genotype of HDL-C level greater than AG, GG genotype.rs708272 TT genotype of HDL-C level greater than CT, CC genotype. rs289714 of CC, CT gene type who HDL-C levels below the TT genotype, above differences were statistically significant(P <0.05). AG genotype LDL-C levels Uighur significantly lower than the control group rs12149545 GG genotype.rs3764261 GT genotype of LDL-C levels were significantly lower than GG genotype. rs711752 of AG, AA genotype TG levels significantly lower than the GG genotype, AG genotype LDL-C level of less than GG genotype, AA genotype HDL-C level higher than GG genotype. rs708272 CT genotype of TG levels below CC gene type person, TT, CT genotype LDL-C levels CC genotype, TT genotype HDL-C levels higher than CC genotype, above differences were statistically significant(P <0.05).3.Uyghur population, exist between rs12149545 and rs3764261, rs711752 and rs708272 complete linkage disequilibrium(D’=1). rs12149545 and rs1800775, rs711752, rs708272, rs289714 loci, rs3764261 and rs1800775, rs711752, rs708272 loci, rs1800775 and rs711752, rs708272 loci, rs1801706 and rs289714,rs5882 bits are having a strong linkage disequilibrium between two loci(D’>0.8 or r2≥0.33). rs3764261 and rs289714, rs1800775 and rs5882, rs1801706, rs1801706 and rs711752, rs708272 locus of between two loci inter are a weak linkage disequilibrium(0.5 <D ’<0.8).4.CETP 8 loci in Uygur population haplotypes constructed 9 species. Including Hap 2-GGCGCCAG,Hap 3-ATAATTAG, Hap 4-ATAATTGG, Hap 7-GGAGCTAG and Hap 8-GGAATTAG haplotype frequencies differences between dyslipidemia and the normal control group was statistically significant(P<0.05).5.Kazakh population CETP gene 10 SNP genotypes were not significantly different in dyslipidemia group and the control group(P> 0.05). Dyslipidemia group except rs1800775 C allele frequency was higher, and the rest minor allele frequency loci were lower than the control group, but all SNP frequencies between the two groups did not differ(P> 0.05).6.Kazakh population dyslipidemia group of rs1800775 CC genotype TG was significantly lower than AA genotype. rs711752 AA genotype of TG, TC, LDL-C levels were higher than GG genotype, AG genotype. The TT genotype of rs708272 TG, TC, LDL-C levels were higher than CT, CC genotype. rs5882 the GG genotype HDL-C levels lower than AA genotype, and above differences were statistically significant(P <0.05). Kazakh control group rs12149545 genotype of AG HDL-C levels higher than GG genotype. rs3764261 GT genotype of HDL-C levels higher than GG genotype. The AA genotype LDL-C levels below rs711752 GG genotype. rs708272 TT genotype of LDL-C levels lower than CC genotype,above differences were statistically significant(P <0.05).7.Kazakh people, there is complete linkage disequilibrium( D’=1) between rs12149545 and rs3764261, rs711752 and rs708272, rs5882 and rs1801706. rs12149545 and rs1800775, rs711752,rs708272 loci, rs3764261 and rs1800775, rs711752, rs708272 locus, rs289714 and rs1801706 between two loci strong linkage disequilibrium(D’> 0.8 or r2≥0.33). rs1800775 and rs711752, rs708272, rs5882 sites have weak linkage disequilibrium between two loci(0.5 <D’ <0.8).8.CETP 8 loci in the Kazakh population haplotypes constructed 10 species. Including differences Hap 6-GGAGCTAG and Hap 9-GGCGCTGG haplotype frequencies between dyslipidemia and the normal control group was statistically significant( P <0.05).Conclusions :1.CETP gene 10 SNP loci, rs12149545, rs3764261, rs1800775, rs711752 and rs708272 five genotypes polymorphism and dyslipidemia risk Uygur Xinjiang related. A allele(rs12149545), T allele gene(rs3764261), A allele(rs711752) and T allele(rs708272) may be protective factors Uighur dyslipidemia, C allele(rs1800775) may be Uighur dyslipidemia risk factors.2.Uygur population, CETP gene rs12149545 and rs3764261 gene polymorphism and LDL-C,HDL-C levels related. rs711752 and rs708272 genotype polymorphisms associated with TG, LDL-C,HDL-C levels, rs1800775 and rs289714 gene polymorphism associated with HDL-C levels.3.Uygur population CETP gene rs12149545 and rs3764261, rs711752 and rs708272 between two loci with full linkage disequilibrium.4.Hap 3-ATAATTAG, Hap 4-ATAATTGG and Hap 7-GGAGCTAG haplotype may be protective factors Uygur dyslipidemia. Hap 2-GGCGCCAG and Hap 8-GGAATTAG haplotype may Uygur dyslipidemia risk factors.5.CETP gene 10 SNP locus genotype polymorphism and Kazakh dyslipidemia risk are irrelevant.6.Kazakh population, CETP gene rs12149545 and rs3764261 gene polymorphism associated with HDL-C levels. rs711752 and rs708272 genotype polymorphisms associated with TG, LDL-C, HDL-C levels. rs1800775 gene polymorphism and TG levels related, rs5882 genotype polymorphisms associated with HDL-C levels.7.Kazakh CETP gene rs12149545 and rs3764261, rs711752 and rs708272 between two loci with full linkage disequilibrium.8.Hap 6-GGAGCTAG and Hap 9-GGCGCGCC haplotype may be Kazakh dyslipidemia risk factors.
